基于信息熵理论和方差分析比较猪牙皂不同组分对小鼠缺血缺氧及血脑屏障通透性影响 被引量:11

摘要 目的:比较猪牙皂中不同组分对小鼠缺血缺氧保护作用及生理病理状态下小鼠血脑屏障通透性影响,探究猪牙皂"通关开窍,醒脑复神"的作用成分。方法:将小鼠随机分为溶媒组、尼莫地平7. 80mg/kg组、猪牙皂总黄酮4. 46mg/kg组、猪牙皂总皂苷17. 55mg/kg组、猪牙皂芳香水130mg/kg组,采用小鼠常压耐缺氧试验,急性脑缺血缺氧试验及正常与病理状态下血脑屏障通透性试验,观察猪牙皂中各组分物质对小鼠的存活时间及生理病理状态下小鼠脑组织中伊文斯兰含量的影响。采用熵权法和方差分析对比猪牙皂中不同组分的作用。结果:与溶媒组比猪牙皂总皂苷17. 55mg/kg、尼莫地平7. 80mg/kg均可以延长小鼠缺氧条件下存活时间,且可以延长小鼠断头后喘息时间;正常生理状态下猪牙皂总黄酮4. 46mg/kg、猪牙皂芳香水130mg/kg能显著提高小鼠脑内EB含量,尼莫地平7. 80mg/kg有提高趋势,总皂苷17. 55mg/kg显著降低小鼠脑组织中EB含量。与模型组比芳香水130mg/kg组能显著提高脑缺氧、缺血模型小鼠脑组织中EB含量。熵权法分析可得:芳香水130mg/kg与总皂苷17. 55mg/kg对小鼠脑缺血缺氧的保护作用在正常与病理状态下血脑屏障通透性的综合影响强于总黄酮4. 46mg/kg。结论:猪牙皂中芳香类成分和皂苷类成分具有较好的"通关开窍,醒脑复神"的作用。 Objective: To compare the different components of Fructus Gleditsiae Abnormalis on the blood-brain barrier permeability of mice under Physiological and pathological state and its protective effects on the Ischemia and hypoxia in mice,then to study the active components which have the effect of ' Tong Guan Kai Qiao,Xing Nao Fu Shen' Methods: Kunming mice were randomly divided into the control group,7. 80mg/kg nimodipine group,4. 46mg/kg total flavonoid group,17. 55mg/kg total saponin group and 130mg/kg Aromatic water group. Through anti-hypoxia experiment,acute cerebral ischemia and hypoxia and Blood-brain barrier permeability test under normal and pathological conditions,effects of different components of Fructus Gleditsiae Abnormalis on mouse survival time and Evans EB content in mouse brain tissue were observed. Then the entropy weight method and variance analysis were used to compare the effects of different components in Fructus Gleditsiae Abnormalis. Results: Compared with the control group,Survival time of mice were Significantly Prolonged after administration of 17. 55mg/kg total saponin and 7. 80mg/kg nimodipine group,mice breathing time after decapitation was significantly prolonged. Under physiological conditions,EB content in mouse brain was significantly increased in 4. 46mg/kg total flavonoid and 130mg/kg aromatic water groups,and was significantly decreased in 17. 55mg/kg total saponin group. Compared with the model group,EB content in the mouse brain was significantly increased in 130mg/kg aromatic water group. The entropy weight method showed that 130mg/kg aromatic water and 17. 55mg/kg total saponin had more significant protective effect than flavonoid on mice with cerebral ischemia and hypoxia and Blood-brain barrier permeability under physiological and pathological conditions. Conclusion: The results explain that aromatic water and total saponins are components in Fructus Gleditsiae Abnormalis that show ' Tong Guan Kai Qiao,Xing Nao Fu Shen' effect.
