芙蓉凝胶对急性乳腺炎大鼠的抗炎作用及机制 被引量:4

摘要 目的考察芙蓉凝胶对LPS诱导急性乳腺炎大鼠乳腺组织MPO表达及炎症因子IL-1β、IL-6、TNF-α水平的影响,并探讨其抗炎作用可能的机制。方法取分娩后雌鼠随机分为空白组、假手术组、模型组、阳性组和芙蓉凝胶低、高剂量组。在大鼠第4对乳腺处,空白组、假手术组、模型组贴空白凝胶,阳性组和芙蓉凝胶高、低剂量组贴相应药物。每天给药1次,连续4 d。第5天,假手术组注入灭菌PBS溶液,模型组、药物组注入LPS溶液,造模后,再给药1次。给药24 h后,取各组大鼠乳腺组织,观察其病理学变化情况,采用免疫组化法检测组织中MPO表达,采用酶联免疫吸附测定法检测组织IL-1β、IL-6、TNF-α的水平,用熵权法计算综合评分,以此评价芙蓉凝胶对急性乳腺炎模型大鼠的综合影响。结果与空白组比较,模型组MPO表达及炎症因子水平升高(P<0.01);与模型组比较,阳性组和芙蓉凝胶高剂量组MPO表达及炎症因子水平均降低(P<0.01)。熵权法计算得阳性组、芙蓉凝胶低剂量组、芙蓉凝胶高剂量组综合评分分别为0.565、0.083、0.352。结论芙蓉凝胶对LPS诱导急性乳腺炎大鼠有一定保护作用,且高剂量组作用强于低剂量组,其作用机制可能与抑制大鼠乳腺组织MPO表达有关。 AIM To investigate the effects of Hibiscus mutabilis gel on the expression of MPO and levels of inflammatory factors IL-1β,IL-6 and TNF-αin mammary gland of rats with LPS-induced acute mastitis,and to explore the possible anti-inflammation mechanism.METHODS Postparturition female rats were randomly divided into blank group,sham-operated group,LPS model group,positive group,and groups for low dose and high dose uses of Hibiscus mutabilis gel.The corresponding administration plan,with the blank gel given to the groups other than the positive group and Hibiscus mutabilis gel groups,was applied to the fourth pair of breast once daily for four consecutive days.On the fifth day,the rats in each group were given the corresponding medicine once again following sterile PBS solution injection to the the sham-operated group and LPS solution injection to the model group and the drug groups.Twenty-four h later,the rats had their mammary tissue removed for observation of the histopathological changes,the detection of MPO expression by immunohistochemistry,and the determination of levels of IL-1β,IL-6 and TNF-αby enzyme-linked immunosorbent assay(ELISA).And overall assessments among different groups due to the influencing gels were conducted by the entropy weight method.RESULTS Compared with the blank group,the model group shared increased MPO expression and the levels of inflammatory factors(P<0.01).Compared with the model group,the positive group,low-dose Hibiscus mutabilis gel group,and high-dose Hibiscus mutabilis gel group displayed decreased MPO expression and levels of inflammatory factors(P<0.01),who shared resultant comprehensive score of 0.565,0.083 and 0.352 from entropy weight method,respectively.CONCLUSION The dose-dependent protective effects of Hibiscus mutabilis gel upon rats with LPS-induced acute mastitis may be associated with the inhibition of MPO.expression.
