摘要
目的:建立HPLC波长切换法同时测定复方桔梗止咳片中6个成分含量的方法。方法:采用Waters XBridge C18色谱柱(250 mm×4.6 mm,5μm),以0.05%磷酸-乙腈为流动相进行梯度洗脱,流速为1.0 ml·min-1,检测波长为326 nm(绿原酸、甘草苷、异绿原酸A、3,6’-二芥子酰基蔗糖、异绿原酸C)、220 nm(款冬酮)。结果:绿原酸、甘草苷、异绿原酸A、3,6’-二芥子酰基蔗糖、异绿原酸C、款冬酮的线性范围分别为18.40~220.74 ng(r=0.999 5)、13.09~157.08 ng(r=0.999 6)、0.34~4.07 ng(r=0.999 3)、9.67~116.04 ng(r=0.999 0)、7.06~84.69 ng(r=0.999 2)、6.85~82.15 ng(r=0.999 2);平均加样回收率(RSD)分别为99.70%(1.41%),100.43%(1.68%),99.03%(2.21%),100.82%(0.45%),100.33%(1.35%),99.23%(1.06%)(n=6)。结论:本文建立的方法可同时测定复方桔梗止咳片中6个成分的含量,方法准确、简便,为复方桔梗止咳片的质量控制提供参考。
Objective:To establish a method for the determination of 6 components in compound Jiegeng Zhike tablets by HPLC with wavelength switching.Methods:The chromatographic separation was performed on a Waters XBridge C18 column(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-0.05%phosphoric acid aqueous solution with gradient elution.The flow rate was 1.0 ml·min-1.The detection wavelengths were set at 326 nm for chlorogenic acid,liquiritin,isochlorogenic acid A,3,6’-disinapoyl sucrose and isochlorogenic acid C,and 220 nm for tussilagone.Results:The linear range of chlorogenic acid,liquiritin,isochlorogenic acid A,3,6’-disinapoyl sucrose,isochlorogenic acid C and tussilagone was 18.40-220.74 ng(r=0.999 5),13.09-157.08 ng(r=0.999 6),0.34-4.07 ng(r=0.999 3),9.67-116.04 ng(r=0.999 0),7.06-84.69 ng(r=0.999 2)and 6.85-82.15 ng(r=0.999 2),respectively.The average recovery and the corresponding RSD was 99.70%(1.41%),100.43%(1.68%),99.03%(2.21%),100.82%(0.45%),100.33%(1.35%)and 99.23%(1.06%)(n=6),respectively.Conclusion:The method is accurate and convenient,which can provide reference for the quality control of compound Jiegeng Zhike tablets.
