中药射干提取物改善豚鼠呼吸道合胞病毒感染后气道炎症的作用研究 被引量:7

摘要 目的:研究射干提取物对呼吸道合胞病毒(RSV)感染豚鼠咳嗽模型气道炎症的治疗作用。方法:将40只豚鼠随机分为4组,每组10只:空白组、模型组、射干组和阳性组,除空白组予等量等渗盐水滴鼻外,其余3组采用烟熏配合RSV滴鼻造模;造模结束后射干组给与射干提取物治疗,阳性组给与肺宁片治疗。通过枸橼酸雾化诱咳比较各组豚鼠的咳嗽次数;采用HE染色法对肺组织形态学检测;血液体液分析仪检测血液中白细胞总数及分类计数;采用LC-MS/MS法MRM监测模式测定肺组织中花生四烯酸含量,探讨射干提取物对RSV感染后咳嗽模型的抗炎作用。结果:肺组织病理学检查结果显示,模型组支气管周围浸润大量炎症细胞,支气管黏膜充血水肿,气管内上皮细胞脱落,纤毛上皮倒伏,提示RSV感染后气道炎症明显改变。模型组中血清白细胞总数、淋巴细胞计数、单核计数、中性粒细胞计数及嗜酸粒细胞计数均无显著差异(P>0.05);枸橼酸雾化诱咳实验中5 min内咳嗽次数和肺组织中花生四烯酸含量显著高于空白组(P<0.01)。与模型相比,射干组和阳性组感染组肺组织炎症细胞浸润程度明显减轻,支干组和阳性组支气管周围炎症减轻。在枸橼酸雾化诱咳实验中5 min内咳嗽次数显著降低(P<0.05),肺组织中花生四烯酸含量显著降低(P<0.05)。结论:射干通过抑制咳嗽时间,减轻肺组织炎症,降低肺组织花生四烯酸含量,发挥止咳作用,相关机制可能是通过阻断花生四烯酸的炎症通路实现的。 Objective:To investigate the effects of Shegan(Belamcanda chinensis)on RSV infection after cough model and explore the relative mechanism.Methods:Forty healthy male guinea pigs were randomly divided into four groups(n=10 per group):the control group,the model group,the Shegan(Belamcanda chinensis)group and the positive group respectively.Cough model of guinea pigs was induced by smoking,intranasal RSV and pulverization of folic acid.After the model was built,the Shegan(Belamcanda chinensis)extract was administrated in the Shegan(Belamcanda chinensis)group.Feining Tablet was administrated in the positive group.Total numbers of white blood cells before and after the administration were evaluated.Morphological changes of lung tissue were observed by hematoxylin-eosin(HE)staining method.Changes of cough time were observed before and after treatment(within 5 min and after 5 min).In addition,the arachidonic acid content in the lung tissue was assayed by high performance liquid chromatography-mass spectrum method.Results:The successful infection of RSV was verified by determining the Hep-2 lesion induced by lung tissue homogenate lesion.The results of lung histopathological examination showed that lots of inflammatory cells were infiltrated around the bronchi,and bronchial mucosal was hyperemia and edema in the control group.There were no significant differences in serum leukocyte count,lymphocyte count,mononuclear count,neutrophil count and eosinophil count in the model group(P>0.05).The number of cough and the content of arachidonic acid in lung tissue were significantly higher than those in the control group(P<0.01).Compared with the model,the degree of inflammatory cell infiltration in the lung tissue of the injection group and the positive group was significantly reducedand the inflammation around the bronchi in the branch group and the positive group was alleviated.In the citrate atomization and cough test,the number of coughing was significantly decreased within 5 min(P<0.05),and the content of arachidonic acid i
