摘要
为了获得本实验室3株(HuB-1、HeB10XS02、SD10LY01)传染性法氏囊病病毒(IBDV)分离毒株的全基因组序列,并了解其序列特征,首先测定了这3株毒株对SPF鸡的致死率,然后利用融合PCR技术扩增获得全基因组序列,对其进行遗传进化分析。结果显示,3株毒株对SPF鸡的致死率均在60%以上,为IBDV超强毒株(vvIBDV)。遗传进化分析表明,3株分离毒株的A节段均位于超强毒分支。IBDV的B节段分为明显的3个亚群,其中HuB-1、SD10LY01的B节段属于第Ⅲ亚群,HeB10XS02的B节段则属于第Ⅱ亚群。第Ⅰ、Ⅱ、Ⅲ亚群之间在VP1蛋白上存在17个保守氨基酸差异位点,其中第145~147位氨基酸形成独特的'三联体'基序,不同亚群具有不同的基序。本研究结果为更好地了解vvIBDV基因组特征以及致病性分子基础的研究提供了重要理论依据。
To determine the sequence characteristics of three infectious bursal disease virus(IBDV)isolates(HuB-1,HeB10XS02 and SD10LY01),firstly,the mortality of these three isolates in SPF chicken was detected and then their genomic sequences were amplified by using fusion PCR.The results showed that all three IBDV isolates could caused the mortality more than 60%and thus they are belongs to very virulent IBDV(vvIBDV).The phylogenetical analyses indicated that their segments A were located in the branch of vvIBDV.The segments B of IBDVs were divided into three distinct subgroups with phylogenetic and analysis,and the segments B of HuB-1and SD10LY01 were located in subgroupⅢ and HeB10XS02was located in subgroupⅡ.A total of 17 conserved amino acid mutations were present among subgroupsⅠ,ⅡandⅢ.A unique'triplet'motif was formed by VP1 145-147 amino acids,and the motifs were different between the subgroups.This study gives us a better understanding of genomic features of vvIBDV and provides the basis for further studying of the pathogenicity of vvIBDV.
