摘要
目的分析吉林省成人麻疹病毒流行株血凝素(Hemagglutinin,H)蛋白抗原位点变异。方法选取吉林省2001-2016年成人麻疹病毒株,经复苏传代、提取核酸、基因扩增获得麻疹病毒H蛋白的分段产物,采用生物信息学软件进行序列分析。结果本研究获得吉林省30株成人麻疹病毒株,与疫苗株S191、H1a基因型代表株China93-4相比,其H蛋白的组间遗传距离分别从2001年的0.050、0.005增加到2016年的0.055、0.011。H蛋白抗原决定簇位点211、240和303发生氨基酸变异,多个位于H蛋白外膜表面的位点发生变异,变异情况与熵值分析结果一致。所有麻疹病毒株H蛋白均保留了前4个糖基化位点,第5个糖基化位点NLS238-240缺失。结论吉林省2001-2016年成人麻疹病毒H蛋白的遗传距离呈逐年增大趋势,部分抗原决定簇位点和外膜表面位点发生氨基酸变异。
Objective To analyze variations of hemagglutinin(H)antigenic epitopes of measles virus strains isolated from adults in Jilin province.Methods We selected measles virus strains f rom Jilin adults during 2001-2016.We obtained H protein f ragments of measles virus by recovering viruses,extracting RNA,and amplifying genes for a genetic sequence analysis using a bio-informatics software.Results We obtained 30 measles virus isolates f rom Jilin adults.The genetic distance of their H proteins increased from 0.050 and 0.005 in 2001 to 0.055 and 0.011 in2016 compared with the S191 vaccine strain and the China 93-4 H1a reference strain.Amino acid variations occurred at sites 211,240,and 303 of the H protein antigenic epitopes and at sites on H protein outer membrane surfaces;these variations matched the entropy analyses.All measles virus strains retained four glycosylation sites;the fifth glycosylation site NLS238-240 was absent.Conclusions Measles virus strains from Jilin adults showed an increasing genetic distance of H proteins f rom 2001 to 2016,with amino acid variations at antigenic epitopes and the outer membrane surface.
