摘要 目的 :观察回转器模拟失重对大鼠骨肉瘤成骨样细胞 (ROS17/ 2 .8)分化相关物质表达的作用 .方法 :对培养的ROS17/ 2 .8细胞采用回转器模拟失重 72h ,同时设 1G对照组 .提取细胞总RNA ,进行反转录PCR ,检测I型胶原 (1链(collagenIα1)和碱性磷酸酶 (ALP)mRNA的表达情况 .同时采用ELISA法测定细胞内I型胶原蛋白的表达量 .结果 :模拟失重 72h后 ,ROS17/ 2 .8细胞内collagenIα1和ALP的基因表达量以及I型胶原蛋白的表达量显著低于对照组 (P <0 .0 5 ) .结论 :回转器模拟失重 72h可使大鼠骨肉瘤成骨样细胞 (ROS17/ 2 .8) AIM: To investigate the effect of rotating clinostat simulated weightlessness on the expression of differentiation related substances in rat osteosarcoma osteoblast like cells (ROS17/2.8). METHODS: One group of cultured ROS17/2.8 cells were exposed to rotating clinostat simulated weightlessness for 72 h. At the same time, another group of cells were cultured in 1G condition as control group. The total RNA in cells was isolated and reverse transcription PCR analysis was performed. The gene expression of α1 chain of type I collagen (collagen Iα1) and alkaline phosphatase (ALP) was examined. The type I collagen protein in cells was quantified by ELISA. RESULTS: After 72 h exposure of ROS17/2.8 cells to simulated weightlessness, the expression of collagen Iα1 and alkaline phosphatase gene and the quantity of type I collagen protein was significantly lower than that in control group ( P <0.05). CONCLUSION: Exposure to rotating clinostat simulated weightlessness for 72 h might reduce the differentiation of rat osteosarcoma osteoblast like cells.
