摘要
目的 :测定Caspase - 3在髓母细胞瘤中的表达水平 ,探讨其在髓母细胞瘤中的作用及其与细胞凋亡的关系。方法 :用免疫组化法测定在 30例髓母细胞瘤以及 10例正常人脑组织中的Caspase- 3蛋白表达 ,慢性扁桃体炎和淋巴结炎标本作阳性对照 ,PBS作一抗为阴性对照。结果 :在 30例髓母细胞瘤中 ,2 4 ( 80 .0 0 % )例不同程度表达Caspase- 3蛋白 ,Caspase- 3蛋白阳性染色见于细胞核、细胞浆 ,或细胞的两个部分。在正常人脑组织中未见到Caspase - 3表达。结论 :Caspase -3与髓母细胞瘤的形成进展、转移和凋亡有关。测定Caspase -
Objective:To measure the caspase-3 expression level in medu ll oblastomas and to probe its presumable roles in the genesis and apoptosis of hum an medulloblastomas. Methods: Expressions of caspase-3 in 10 contr o l specimens obtained from patients, for them the mental and nervous diseases had been eliminated and in 30 human medulloblastomas were examined by routine immunohist och emical procedures. The specimens of chronic tonsillitis and lymphonoditis were u sed as positive control and specimens with PBS as first antibody were used as negative control. Results:Of the 30 medulloblastomas, 24(80.00%) patients expressed caspase-3 protein, in nuclear or cytoplasmic or in both cellular com part ments at variable levels. Caspase-3 immunoreactivity was not found in controlled br ain specimens. Conclusions: Apoptosis-cell programmed death-is s t rictly controlled by a crucial enzyme: Caspase, (Cysteinyl-aspartate specific p roteinase, apoptosis executioner), which is tigh tly involved not only in the genesis, progression and metastasis of medulloblast omas, but in apoptosis of the neoplasmas as well , and therefore the determina t ion of caspase-3 is of clinical value for assessing the tumor biological behavio r and patient′s prognosis.
