四氧嘧啶剂量和给药途径对制作大鼠糖尿病模型的影响 被引量:32

EFFECTS OF COPYING DIABETES MELLITUS RAT MODEL BY DIFFERENT DOSES AND INJECTING METHODS WITH ALLOXAN

摘要 目的 探索四氧嘧啶制作大鼠糖尿病模型所需的最低剂量和不同给药途径对制作大鼠糖尿病模型的影响。方法 用四氧嘧啶 3种剂量于雌雄各半的SD大鼠尾静脉注射和腹腔注射制作糖尿病模型。连续 15d测定有关指标 (血糖、尿糖、尿量、饮水量、体重 )。结果 ①静脉给药组动物有关指标变化比腹腔给药组明显 ;,②静脉给药组低剂量组模型失败 ,高、中剂量组模型制作成功。结论 ①用四氧嘧啶制作大鼠糖尿病模型静脉给药优于腹腔给药 ,②用四氧嘧啶以静脉给药方法成功制作大鼠糖尿病模型未禁食情况下需剂量≥ 4 0mg/kg。 Objective To explore the effects of minimum dose and different injecting methods in copyint diabetes mellitus rat model with alloxan(ALT). Methods Sixty rats were divided into two groups and three different doses of alloxan were given to each group by caudal vein injecting or abdominal cavity injecting methods. After that, the haematic glucose, urine glucose, urine quantity, drinking water quantity and body weight of rats were examined for successive 15 days. Results (1) The changes of parameter are more significant in caudal vein injecting group than in abdominal cavity injecting group. (2) In caudal vein injecting group, rats injected with low dose were end in failure but rats injected with medium or high doses were success in copying the model. Conclusion (1) For copying diabetes mellitus rat model, the caudal vein injecting is much better than abdominal cavity injecting. (2)The minimum dose of ALT for copying the model by caudal vein injecting is ≥40 mg/kg.
