摘要
目的:探讨IL-37对肝细胞癌的MHCC97H细胞CD133表达的影响。方法:肝癌细胞系MHCC97H细胞分为对照组和过表达IL-37组,利用lip3000试剂盒进行基因转染,PCR检测IL-37及CD133的表达。结果:过表达组IL-37m RNA上升了37%(P<0.05),显示成功过表达IL-37基因。过表达组CD133 m RNA下降了48%(P<0.05),显示过表达的IL-37抑制了CD133的m RNA表达。与对照组比较,过表达IL-37组细胞形态变为圆形,细胞体积缩小,细胞的数量也减少。结论:IL-37过表达抑制MHCC97H细胞的CD133的m RNA表达及细胞生长。
Objective To investigate the effect of IL-37 on the expression of CD133 in MHCC97H cells of hepatocellular carcinoma(HCC). Methods Hepatocarcinoma cell line MHCC97H cells were divided into control group and IL-37 group. Gene transfection was performed using lip3000 kit. The m RNA expression of IL-37 and CD133 was detected by PCR. Results The expression of IL-37 m RNA in the IL-37 group increased by 37%(p<0.05), indicating that the IL-37 gene was successfully over expressed. CD133 m RNA of IL-37 group decreased by 48%(p<0.05), indicating that over expression of IL-37 inhibited CD133 m RNA expression. Compared with the control group, cells of IL-37 group became rounded. The cell volume the number of cells decreased. Conclusion IL-37 over expression inhibits the expression of CD133 m RNA and cell growth in MHCC97H cells.
