小鼠神经管发生中程序性细胞死亡的时空规律 被引量:1

摘要 目的:研究小鼠神经管发生中程序性细胞死亡(programmedcelldeath,PCD)的时空规律。方法:分别取交配后9.0d、交配后9.5d、交配后10.5d鼠胚做石蜡包埋连续切片的TUNEL染色。进行神经上皮凋亡细胞的计数分析。结果:观察到交配后9.0d神经上皮的细胞凋亡率(0.1815±0.14)最高,交配后9.5d后细胞凋亡率(0.1377±0.09)降低,交配后10.5d(0.1284±0.07)降至更低。从空间分布看,神经管头端的凋亡细胞明显多于尾端。结论:交配后9.0d神经管的高细胞凋亡率,可能与神经管的关闭有关;神经管颅侧的细胞死亡与脑泡的塑形有关。 AIM:To study the spatio temporal distributions of programmed cell death(PCD) in the development of mouse neural tube. METHODS:The mouse embryos were taken at E 9.0 d,E 9.5 d,and E 10.5 drespectively,and were performed by preparation for paraffin series section using the staining of terminal deoxynucleotidyl transferase mediated dUTP biotin nick end labeling(TUNEL).The apoptotic cell number of neuroepithelium was accounted and analysed in present study. RESULTS:It was found that there were a peak of cell apoptosis rate (0.181 5±0.14)appearing at E 9.0 d.Then the rate of cell apoptosis was decreased to (0.137 7±0.09) at E 9.5 d and continually declined at E 10.5 d(0.128 4±0.07).In the spatial distribution,the number of apoptotic cells was observed in anterior part of the neural tube more than that in caudal part. CONCLUSION:The high rate of cell apoptosis may be related with the closure of neural tube at E 9.0 d,while the cell death in the cranial neural tube is related with the remodel of the brain vesicle.
