摘要
目的 :研究bcl- 2和细胞凋亡在多形性腺瘤 (PA)和恶性多形性腺瘤 (MPA)发生中的作用。方法 :选取PA30例 ,MPA10例和正常腮腺组织 10例为研究对象 ,进行bcl- 2原位杂交染色和细胞凋亡的TUNEL染色。用图像分析仪对bcl- 2的染色结果进行定量分析 ,并计算细胞凋亡指数 ,最后进行统计学分析。结果 :bcl- 2mRNA染色的阳性信号主要表达在腺管样的上皮细胞中 ,正常组和PA组之间无显著性差异 (P >0 .0 5 ) ,正常组和MPA组、PA组和MPA组之间均有显著性差异 (P <0 .0 5 )。凋亡细胞散在分布于腺管样的上皮细胞中。PA组的凋亡指数明显高于MPA组 (P <0 .0 5 )。bcl- 2的阳性表达强度与凋亡指数呈显著的负相关。结论 :bcl -
Objective:To study the effects of bcl-2 and apoptosis on the pathogenesis of pleomorphic adenoma (PA) and malignant pleomorphic adenoma (MPA).Methods:Normal and pathological samples were analyzed including 30 cases of PA, 10 cases of MPA and 10 cases of normal parotid glands. Expression of bcl-2 mRNA and apoptosis were performed by in situ hybridization and TUNEL staining. Expression of bcl-2 was quantified by image analysis instrument and apoptosis index was calculated. The data was analyzed by statistics method.Results:bcl-2 mRNA mainly expressed in the tubular epithelial cells. There was no significant difference between normal group and PA group (P<0.05).Significant differences were found between normal group and MPA group, PA group and MPA group.Apoptosis cells mainly scattered in ductlike epithelial cells. Apoptosis index was significant higher in PA group than that in MPA group (P<0.05). There was a significant passive correlation between bcl-2 mRNA and apoptosis index.Conclusions:The occurrence and malignant transformation PA were closely related with the decrease of bcl-2 expression and apoptosis index.
