p73及PCNA在腮腺良、恶性多形性腺瘤组织中的定位与表达

摘要 目的通过检测来源腮腺的良性、恶性多形性腺瘤组织中的p73、PCNA的定位与表达的情况,探索其与腮腺的良、恶性多形性腺瘤之间的关系及机理。方法收集已确诊的来源于腮腺的良、恶性多形性腺瘤石蜡标本共40例(其中肿瘤良性25例,恶性15例);另制备明确为来源正常腮腺组织的标本20例作其对照。采取S-P免疫组织化的方法进行检测。结果 p73在良性多形性腺瘤阳性表达率显著高于恶性多形性腺瘤(P<0.01),其差异有统计学意义。PCNA的阳性表达率在健康腮腺组织中的明显低于在腮腺来源的良、恶性多形性腺瘤组织组(P<0.01),其差异有统计学意义。结论 p73与腮腺的良、恶性多形性腺瘤的发生、发展关系密切,PCNA过度表达与腮腺的良、恶性多形性腺瘤的发生与发展有关联,检测p73、PCNA可探索癌前状态的本质,对腮腺多形性腺瘤的预后及良、恶性的鉴别诊断有重要意义。 Objective To detect the localization and expressions of protein P73 and PCNA in the tissues of benign and malignant pleomorphic adenoma in parotid gland and to explore the relationship and mecha- nism between the P73 and benign and malignant pleomorphic adenoma in parotid gland. Methods The par- affin specimens of 40 cases of confirmed benign and malignant pleomorphic adenoma (25 benign cases and 15 malignant cases) and another 20 normal cases were used as controls were collected and detected through S-P immunohistochemical method. Results The positive expression rate of P73 in the in the tissues of be- nign pleomorphic adenoma was significantly higher than that in the tissues of malignant pleomorphic ade- noma ( P 〈0.01). The positive expression rate of PCNA in normal cases was significantly lower than that of benign and malignant pleomorphie adenoma ( P 〈0.01). Conclusion P73 may be closely related to the occurrence and development of benign and malignant tumors of the parotid gland. Over-expressions of PC- NA may be associated with the occurrence and development of benign and malignant tumors of the parotid gland. Detecting p73 and PCNA can be used to study the nature of the precancerous state to guide the clin- ical treatment and it has important significance for the prognosis and differential diagnosis of benign andmalignant tumors of the parotid gland.
