摘要
目的:了解铜绿假单胞菌对亚胺培南的耐药机制。方法:选取临床分离的铜绿假单胞菌89株,采用纸片扩散法行药敏试验、紫外线吸收法行膜微孔蛋白opr D2基因测定、纸片协同试验测定金属β-内酰胺酶,同时采用Etest试验了解氰氯苯腙对亚胺培南最低抑菌浓度的影响。对所有试验结果进行统计分析。结果:耐药铜绿假单胞菌菌株opr D2含量明显低于敏感铜绿假单胞菌菌株(P<0.05);且有耐药铜绿假单胞菌菌株产生β-内酰胺酶;同时还有耐药铜绿假单胞菌菌株存在主动外排泵出机制。结论:opr D2含量降低、β-内酰胺酶产生、主动外排泵出机制是导致铜绿假单胞菌对亚胺培南耐药的主要作用机制。
Objective:To understand the mechanism of imipenem resistant pseudomonas aeruginosa.Methods:89 strains of clinical isolates of pseudomonas aeruginosa were selected.Disk diffusion method was used for drug sensitivity test,ultraviolet absorption method for the determination of microporous membrane protein oprD2 gene,disk synergy test determination ofβ-metal enzyme, while understanding the effect of cyanide chlorophenylhydrazone on imipenem minimum inhibitory concentration by using Etest test.Results:oprD2 resistant pseudomonas aeruginosa strains were significantly lower than the sensitive strain of pseudomonas aeruginosa(P<0.05),and drug resistant pseudomonas aeruginosa strains produceβ-metal enzyme;at the same time there are drug resistant pseudomonas aeruginosa strains existactive efflux pump mechanism.Conclusion:Decreased the content of oprD2,β-metal enzyme producing,active efflux pumpmechanism were the main mechanism resulting in pseudomonas aeruginosa resistantto imipenem.
