摘要
单细胞核糖核酸(RNA)测序是全面研究免疫细胞及其功能的高效技术平台,近年获得了快速的发展。本文就该技术近期在艾滋病免疫学方面的重点研究进展进行综述:主要包括对艾滋病病毒(HIV)感染的CD4^+T淋巴细胞和HIV感染后的潜伏细胞及其特征和相关机制的研究;此外,还有对精英控制病人淋巴组织内的CD8^+T淋巴细胞以及树突状细胞的分析研究,发现了与HIV的有效控制密切相关的细胞亚群和转录特征;同时还讨论了下一步研究重点和方向。
Single-cell RNA sequence(sc-RNA-seq)technology enables analysis of characterization and function of single cell accurately,and has been developed and applied rapidly in recent years.This article aims to review the research on HIV immunity with this technology,including the discovery of HIV permissive CD4^+T cells and the characteristics and maintenance mechanisms of HIV latent reservoir,as well as the state of latency cell after reactivation.In addition,HIV specific resident memory CD8^+T cells in lymphoid tissues and dendritic cells(DCs)cells in PBMC,which help for effective control of HIV in HIV elite controllers,has also been researched.These cells are the main targets of HIV immunology research,and there are a lot of new knowledge in the studies,which is worthy of further study and exploration.
