摘要 目的:探讨三磷酸腺苷生物发光法(ATP鄄BLA法)在白血病细胞药敏实验中的可行性。方法:采用ATP鄄BLA法,用液体闪烁记数仪检测ATP标准品以及白血病细胞株K562在不同细胞浓度、药物浓度、培养时间下的荧光发光值。结果:ATP标准品浓度、K562细胞浓度的对数与荧光发光值的对数具极好的线性关系(P<0.001);药敏实验时,结束细胞培养的时间以第4d为宜,药物浓度的选择以1×PPC为最佳。结论:ATP鄄BLA法在白血病药敏试验中是可行的,有一定的临床应用价值,值得进一步研究。 Objective: To Study the efficiency of ATP-Bioluminescence Assay (ATP-BLA) in chemosensitivity test of leukemic cell. Methods: Under ATP-BLA, multilabel counter was used to determine the luminescent value of ATP and leukemic cell line K562 which had different cellular concentration, cultured time and drug-exposed conentration. Results: There is a wonderful linear relationship between the log concentration of ATP, K562 and log luminescent value (P < 0.001); For chemosensitivity test, the optimal cell-cultured time was in the fourth day and the standard drug-exposed concentration was 1×PPC. Conclusion: ATP-BLA is a highly efficient and reliable assay for the chemosensitivity test of leukemia cell and worthy for further clinical investigation.
