摘要
目的 探讨衰老对肺泡Ⅱ型细胞合成表面活性物质代谢的影响。方法 利用形态计量和Northern杂交方法比较了幼(1~2月龄)、中(9~10月龄)和老年(25~28月龄)大鼠肺泡Ⅱ型细胞内板层体数及肺组织内肺泡表面活性物质相关蛋白(SP)A、B和C基因的表达。结果 老年鼠肺泡Ⅱ型细胞内板层体数为12.20±4.99个/细胞,高于幼年鼠的11.25±3.29个/细胞(P<0.01),但低于中年鼠的13.37±4.55个/细胞(P<0.05)。肺组织中SP-A、SP-B和SP-C基因的表达各年龄组之间则无差别。结论 肺泡Ⅱ型细胞对表面活性物质代谢的年龄相关性变化能力可能是代偿性分泌增加而达到的新的动态平衡的结果,并不是老化时肺功能下降的原因。
Objective To investigate effects of the aging on the metabolism of surfactant in alveolar type Ⅱ cells. Methods Morphometric analysis and Northern blot were used to study the numbers of the lamellar bodies of alveolar type Ⅱ cells and the expression of surfactant associated protein(SP) A, B and C genes in lung tissues from young (1-2 months),adult (9-10 months) and old (25-28 months)rats. Results The numbers of lamellar bodies in alveolar type Ⅱ cells from old rats were 12.20 ±4.99 per cell,which was higher than that in young (11.25 ±3.29, P<0.01 )but lower than that in adult rats( 13.37±4.55, P<0.05). The expression of SP-A, B and C genes in lung tissues was not different dramatically from each other. Conclusion Age-related changes of surfactant metabolism in the alveolar type Ⅱ cells may be the results of increasing secretion of the surfactant and are not the causes of decline in lung function of aged rats.
