摘要
目的 探讨应用口服补硒方法纠正慢性肝病患者外周血单个核细胞 (PBMC)免疫功能紊乱的临床价值。方法 患者口服补硒 (2 0 0~ 30 0 μg·d-1) 8~ 12周后分离PBMC ,分别对比观察硒作用前后PBMC膜流动性、白细胞介素 2受体 (IL 2R)表达及其分泌IL 2活性、过氧化脂质丙二醛 (MDA)含量的变化 ,并结合体外PBMC培养实验结果 ,探讨其可能机制。结果 口服补硒治疗后 ,患者PBMC膜流动性、IL 2R表达及其分泌IL 2活性均显著提高 ,血中LPO含量显著减少 ,与未补硒组比较差别均有显著性意义 (P <0 .0 5 ;P <0 .0 1)。体外培养患者的PBMC外加硒作用后上述指标呈相似变化。结论 口服补硒可纠正慢性肝病患者PBMCIL 2系统的功能紊乱 ,其机制可能与抑制PBMC膜的脂质过氧化损伤 。
Objective To study the effects of selenium on the peripheral blood mononuclear cells (PBMC) membrane fluidity and the function of patients with chronic hepatitis. Methods PBMC pretreated with selenium (1.156×10 -7mol·L -1) for 6 hours in vivo or extracted directly from the patients after taking Se-yeast continuously for 8~12 weeks (200 μg of selenium one day) were exposed to Con-A for 48 hours. The membrane fluidity, interleukin-2 (IL-2) productionand interleukin-2 receptor (IL-2R) expression of PBMC and the malondialdehyde (MDA) concentration in medium and plasma were determined. Results The PBMC membrane fluidity, IL-2 production and IL-2R expression of the patients with chronic hepatitis were significantly decreased and the MDA concentration in medium or plasma were significantly increased (P<0.01). Both in vivo and vitro,administration of selenium could reverse the parameters above (P<0.05).Conclusion Supplementing selenium can suppress the membrane lipid peroxidation of PBMC, maintain the membrane fluidity and improve the PBMC function of patients with chronic hepatitis.
