摘要
目的 :研究静态机械拉伸应变对体外培养的人牙周膜成纤维细胞前列腺素E2 (PGE2 )分泌量的影响。方法 :实验的拉伸应变量设为 0 %、8%、12 %、16%、2 0 % 5种 ,加力时间为 2 4、48、72h3种 ,总共分为 15组 ,每组设 3个样本。加力完毕后 ,用RIAELISA法分析每个样本培养基中PGE2 的含量并进行统计分析。结果 :在0 %力值组 ,PGE2 的每天分泌量与加力时间无关 ;在 8%、12 %、16%力值组 ,PGE2 的每天分泌量随着加力时间和力值的增加而递增 ,但每天递增量随着加力时间的增加而减少 ;在 2 0 %力值组 ,PGE2 的每天分泌量与加力时间成反比 ;在各个时间组 ,PGE2 的每天分泌量均在 16%力值处达到最高值。结论 :在细胞的生理应变承受范围 (0 %、8%、12 %、16% )内 ,静态机械拉伸应变促进人牙周膜成纤维细胞PGE2 的分泌 ,但刺激效应随着加力时间的增加而减少 ;当拉伸应变量超过了细胞的生理应变承受范围 (2 0 % ) ,则PGE2 的分泌量会随着加力时间的增加而减少。
Objective: To study the effects of static tensile strai n on the secretion of prostaglandin E 2 (PGE 2) of human periodontal ligament fibroblasts (PLFs). Methods: Human PLFs were treated by tensile strain values of 0%,8%,12%,16% and 20% for 24, 48 and 72 h respecti vely with a self-devised loading apparatus in vitro. All samples were rando mly allocated into 15 groups and there were 3 in each group. After treatment cu lture media of the samples were collected and the content of PGE 2 in each medi um sample was determined using RIA ELISA. The data analysis was carried out with SPSS using Dunnett test. Results: In group of 0% the sec retion of PGE 2 by PLFs per day had no relation with loading time; E 2 secre tion increased with the increace of loading time and with the tensile strain va lue in the group of 8%,12% and 16%(P<0.05). The secretive amount of PGE 2 was inversely proportional to the loading time in the group of 20%. When the cells were under the tensile strain value of 16% the secretion amount of PG E 2 reached the maximum. Conclusion: The static tensile strain of physiological extent (0%,8%,12% and 16%) stimulates the secretion of PGE 2 by human PLFs. When the strain is over the physiological extent(20%) i t will result in the decrease PGE 2 secretion.
