摘要
目的 :以化学去细胞兔神经移植修复大鼠坐骨神经缺损 ,从而探索化学去细胞异种神经移植修复神经缺损的可能性 ;方法 :以 3 .0 %TritonX 1 0 0和 4 .0 %脱氧胆酸钠顺序处理新鲜取材的兔神经 (直径 1 .5mm左右 ) ,移植修复成年wistar大鼠 1 .0cm坐骨神经缺损 ,4个月后行电生理及组织学检查 ,观察神经再生情况 ;结果 :移植神经未被宿主排斥 ,大量再生的神经纤维长过移植物 ,并恢复电传导功能 ;结论 :化学去细胞异种神经可以作为修复周围神经缺损的一种很有前途的方法。
Objective:To observe the nerve regeneration in the adult rats,which had been made a 1.0cm long gap in the continuity of the sciatic nerve and the gap was repaired by rabbit nerves made acellular by chemical extraction.Methods:The sciatic nerves on the left side of 12 rats were exposed and 1.0cm long segments of the nerves were removed and replaced by rabbit nerves decellulated by chemical extraction.After 4 months,the nerve regeneration and electric conductiong function were revealed by morphological studies and electrophysiological tests.Results:In the rats's nerve repaired by acellular nerves,regenerated axons and migrated Schwann cells reoccupied the empty basal lamina tubes of the acellular segments without excessive sign of inflammation following implantation,and stimuli(1.5mA,0.1ms,1.0Hz) to the sciatic nerves proximal to the implanted segment resulted in motor evoked potentials recorded from posterior tibial muscles.Conclusions:Chemical acellular xenogeneous nerve could sustain peripheral nerves regeneration without notabal immune exclusion,therefore they maybe a promising explant to repair nerve defect in clinic.
