The effect of human serum albumin on bacterial adhesion to cardiovascular biomaterials in vitro

摘要 目的 观察人血清白蛋白 (HSA)对涤纶 (Dacron)和膨体聚四氟乙烯 (PTFE)细菌黏附的影响 ,探讨其在心血管生物材料细菌黏附中的作用 ,为临床防治心血管生物材料植入后感染提供有效的方法。 方法 根据生物材料处理的方式不同分为两组 ,实验组 :用 HSA预处理涤纶和 PTFE;对照组 :用生理盐水预处理。使用 3H-胸腺嘧啶脱氧核苷 (3H- TDR)标记的金黄色葡萄球菌、表皮葡萄球菌 ,对两组的涤纶和 PTFE进行动态细菌黏附实验 ,使用β-液闪测量仪 (β- L SC)、扫描电子显微镜 (SEM)观察材料上细菌黏附情况。 结果 实验组金黄色葡萄球菌和表皮葡萄球菌对涤纶和 PTFE的黏附均较对照组少 (P<0 .0 5 )。 结论 用 HSA预处理心血管生物材料可抑制某些细菌的黏附。 Objective To observe the influences of human serum albumin (HSA) on bacterial adhesion to cardiovascular biomaterials of Dacron and polytetrafluoroethylene (PTFE) and provide an effective method for preventing infection of implanted biomaterial. Methods According to various pretreating methods, biomaterials were divided into two groups.Trial group: Dacron and PTFE were preincubated in HSA, control group: Dacron and PTFE were preincubated in physiological saline solution. Staphylococcus aureus and Staphylococcus epidermidis were radiolabeled with tritiated thymidine (3H-TDR), and then started the experiment of bacterial adhesion to Dacron and PTFE in vitro. The radioactivity associated with segments was measured with beta liquid scintillation counting(β-LSC). Scanning electron microscopy (SEM) was also used to observe the phenomenon of bacterial adhesion to biomaterials. Results Compared with control group, segments of Dacron and PTFE coated in HSA decreased the binding of Staphylococcus aureus and Staphylococcus epidermidis to both Dacron and PTFE in trial group (P<0.05). Conclusion Cardiovascular biomaterials coated in HSA can effectively inhibit some bacterial adhesion to cardiovascular biomaterials.
