摘要
目的 研究非小细胞肺癌组织中survivin和P34cdc2 的表达 ,探讨survivin和P34cdc2 的表达与非小细胞肺癌临床病理特征的关系。方法 随机收集非小细胞肺癌 (含癌旁细支气管和 /或小支气管增生组织和正常肺组织 )标本 10 0例。采用免疫组织化学SP法染色。结果 在癌组织与癌旁细支气管和 /或小支气管上皮增生组织及正常肺组织中survivin和P34cdc2 表达差异有显著性 (P <0 .0 1)。在癌组织中有过表达 ,在癌旁细支气管和 /或小支气管上皮增生组织中的表达较正常肺组织中的表达增强 ;癌组织中survivin和P34cdc2 表达呈正相关 (P <0 .0 1) ,癌旁细支气管和 /或小支气管上皮增生组织中 ,survivin和P34cdc2 的表达也呈正相关 (P <0 .0 1)。癌组织类型、分化程度和淋巴结转移与survivin和P34cdc2 的表达均无相关性 (P >0 .0 5 )。不同临床分期的非小细胞肺癌 ,其survivin和P34cdc2 的表达差异有显著性 (P <0 .0 5 )。结论 survivin和P34cdc2 在非小细胞肺癌中有过表达现象 ,在非小细胞肺癌的G2 /M期 ,P34cdc2 可能通过将survivinThr (34)磷酸化而激活或增强细胞的抗凋亡能力 ,从而将细胞周期进程与凋亡调控有机地联系起来。过表达的survivin和P34cdc2 可作为反映非小细胞肺癌细胞分裂增殖、凋亡抑制 (细胞生存?
? Objective To study the expression of survivin andP34 cdc2in human non-small cell lung cancer (NSCLC) and the relationshipbetween such expression and clinicopathological features of NSCLC.Method 100 NSCLC cases with neighboring noncancerous tissue and normal lung tissue were selected at random. These samples were examined by immunohistochemical SP methods.Result The expressions of survivin and P34 cdc2showed significant differences (P<0.01) between cancer tissues, neighboring noncancerous tissues with proliferating epithelial cells of bronchioles and small bronchi, and normal lung tissues. There was overexpression of survivin and P34 cdc2 in NSCLC. The expressions of survivin and P34 cdc2 in neighboring noncancerous tissues with proliferating epithelial cells of bronchioles and small bronchiwere stronger than those in normal lung tissues. A significant positive correlation was found between the expressions of survivin andP34 cdc2 in NSCLC (P<0.01).The positive correlationwas also found between theexpressions of survivin andP34 cdc2in neighboring noncancerous tissues (P<0.01). No statistical significance was found between the different histological types, the differentiation degrees, the lymphatic metastasis and the expressions of survivinandP34 cdc2 (P>0.05). However, statistical difference was marked betweenthe different clinical stages of NSCLC and the expressionsof survivin and P34 cdc2 (P<0.05). Conclusion The overexpression of survivin and P34 cdc2 was found in NSCLC. P34 cdc2may activate or enhance the ability of antiapoptosis by phosphorylating Thr (34) of survivin in NSCLC cells during the G 2/M phase, so that the progression of the cell cycle can be efficiently linkedwith the control of apoptosis by the interaction of the overexpressionsofsurvivin and P34 cdc2 . The overexpression ofsurvivin and P34 cdc2 may be used as marker in showing the dividing and proliferating ability, and the apoptosis-inhibiting ability(lengthening cell lifespan) of NSCL
