摘要
目的:应用硝酸镓进行预防性处理,观察硝酸镓对去卵巢引起的大鼠骨质疏松的预防作用。方法:通过去除大鼠卵巢建立骨质疏松模型。将8月龄雌性SD大鼠(体质量220~270g)单纯随机分为对照组(14只)、去卵巢组(13只)、单纯预防组(13只)和去卵巢预防组(17只)。去卵巢组和去卵巢预防组切除双侧卵巢,对照组和单纯预防组行假手术。对照组和去卵巢组腹腔注射生理盐水1mL/kg;去卵巢预防组和单纯预防组腹腔注射硝酸镓2mL/kg,均为3次/周。12周后收集24h尿液,取血和双侧股骨。测定血、尿、骨镓含量及骨钙含量、骨密度、骨强度。结果:12周后,单纯预防组和去卵巢预防组骨镓含量(μg/g)显著高于对照组和去卵巢组(6.82±0.67,7.56±0.53,0.48±0.11,0.26±0.15;t=-29.72~22.64,P<0.01);去卵巢预防组骨钙含量(17.5±4.0)mg/g、骨密度(0.55±0.06)g/cm2、最大负荷(9.7±0.6)N、最大形变(1.64±0.16)mm均显著高于去卵巢组(t=-2.26,2.62,3.01,3.56,P<0.05),与对照组无显著性差异。结论:硝酸镓对去卵巢引起的大鼠骨质疏松有一定程度的预防作用。
AIM:To prove the preventive function by adopting the Gallium nitrate (GaN) ag ainst osteoporosis caused by ovariectomization of the rats. METHODS:Osteoporotic models of rats were set up by ovariectomizing. The eight -month-old female SD rats (weight:220-270 g) were divided into four groups ra ndomly:Control group (CG, 14 rats),Control ovariectomized group(COG, 13 rats), G a-preventive sham operated group (PSOG, 13 rats), Ga prevent ovariectomized sub -group(POSG, 17 rats).COG and POSG were ovariectomized bilaterally.1 mL/kg Sali na was injected into abdominal cavities in CG, COG and 2 mg/kg GaN in PG, PSOG 3 times a week. After 12 weeks, 24 hours urine was collected,blood and their bila teral femurs were drawn for testing.The contents of blood,urine,bone Calcium, ga llium and bone mineral density (BMD), bone intensity (BI) were measured. RESULTS:After 12 weeks, the contents (μg/g) of bone Ga in POSG were significa ntly higher than that in COG and CG (6.82±0.67,7.56±0.53,0.48±0.11,0.26±0 .15;t=-29.72-22.64,P< 0.01), the contents of bone Calcium(17.5±4.0)mg/g,BMD ( 0.55±0.06)g/cm2, Maximum burthen(9.7±0.6)N, Maximum distortion(1.64±0.16)mm were significantly higher than that of COG (P< 0.05) and had no significant diff erence with that of CG.
