丹参对大鼠心肌缺血再灌注损伤后血浆一氧化氮和心肌c-fos mRNA表达的作用 被引量:27

摘要 目的 探讨丹参 (SM)对大鼠心肌缺血再灌注损伤 (IRI)后血浆一氧化氮 (NO)和心肌c fosmRNA表达等的作用。方法 采用结扎左冠状动脉前降支 30min ,再灌注复制大鼠心肌IRI的模型 ,结扎前 10min注射丹参 ,检测心肌再灌注 4 0min时血浆NO、内皮素 (ET)的含量和心肌组织乳酸脱氢酶 (LDH)、磷酸肌酸激酶 (CK) ,并用原位杂交方法观测左室心肌c fosmRNA的表达。结果 SM组 ( 4 g/kg)较IRI组 ,能显著减少心肌酶的漏出 [LDH :( 10 92± 5 7)U/Lvs ( 1977± 6 4 )U/L ;CK :( 185± 6 )U/Lvs ( 2 0 6± 4 )U/L ,(均为P <0 0 1) ];明显提高血浆NO含量 [( 5 8 3± 4 2 ) μmol/Lvs ( 2 1 8± 2 7) μmol/L ,(P <0 0 1) ],减少ET的生成 [( 2 2 8± 10 ) pg/mlvs ( 313± 13) pg/ml,(P <0 0 1) ],血浆NO与ET比值也明显升高 [( 0 30± 0 0 2 )vs ( 0 0 7± 0 0 1) ,(P <0 0 1) ];IRI组心肌组织c fosmRNA表达的光密度值为 0 2 1± 0 0 3,而SM组光密度值为 0 12± 0 0 3,c fosmRNA的表达明显减少(P <0 0 5 )。结论 SM对心肌的保护作用机制之一与提高血浆NO含量、减少ET的生成 ,下调c Objective To study the effects of Slavia miltiorrhizae (SM) on nitric oxide of plasm and c fos mRNA expression of myocardium induced by myocardial ischemia reperfusion injury (IRI) in rats. Methods Ischemia reperfusion model was induced by left descending anterior coronary artery (LAD) ligation for 30 min, followed by 40 min of reperfusion. After the mice was pretreated with SM, the levels of lactate dehydrogenase (LDH), creatine kinase (CK) in myocardium and the contents of nitric oxide (NO), endothelin (ET) in plasma were determined, and the expression of c fos mRNA in left ventricle myocardium was detected by in situ hybridization.Results As compared with IRI group, SM (4 g/kg) could markedly decrease the enzyme levels of myocardium leakage \[LDH: (1092±57) U/L vs (1977±64) U/L, ( P <0 01); CK: (185±6) U/L vs (206±4) U/L, ( P <0 01)\]; increase the content of NO in plasm \[(58 3±4 2) μmol/L vs (21 8±2 7) μmol/L, ( P <0 01)\], and reduce the level of ET \[(228±10) pg/ml vs (313±13) pg/ml, ( P <0 01)\], and raise the ratio of NO/ET incidentally \[(0 30±0 02) vs (0 07±0 01), ( P <0 01)\]. The c fos mRNA expression in IRI group was (0 21±0 03), and SM obviously decreased the expression of c fos mRNA in myocardium \[optical density (OD): 0 12±0 03 vs 0 21±0 03 in IRI group, P <0 05\]. Conclusion SM could obviously protect myocardial reperfusion injury, and the effects may be associated with the increased content of NO, decreased level of ET in plasma, downregulation of the expression of c fos mRNA.
