摘要
香蕉花叶心腐病由黄瓜花叶病毒(CMV)引起,它有一个三分体正单链RNA基因组。从不同模板制备方法、RT和PCR过程引物浓度的选择等方面,用两步法RT-PCR对香蕉花叶心腐病的检测技术进行了研究。为了简化检测程序,在两步法RT-PCR检测程序的基础上,进一步研究香蕉CMV一步法RT-PCR检测技术,并对两种方法的检测灵敏度进行了比较。结果发现,两种方法都能从相当于100ng的香蕉叶组织中扩增到病毒带,但一步法RT-PCR的灵敏度比两步法略有提高。
Banana mosaic disease is caused by cucumber mosaic virus(CMV), which posses a tripartite, positive-sense, single-stranded RNA genome. Two-step RT-PCR assay has been developed for detection of CMV through comparison of different protocols in RT-PCR. In order to simplify the procedures of the RT-PCR, one-step protocol was developed in this study to investigate CMV in banana tissue by following the two-step RT-PCR protocol. The results showed that CMV within 100ng banana infected tissue could be detected by both of one-step and two-step RT-PCR, and susceptibility of the later was slightly higher than the former.
