摘要
应用香蕉束顶病毒(BBTV)的单克隆抗体(McAb)建立了检测香蕉束顶病的间接ELISA方法,其检测香蕉病组织汁液的最大稀释度为1:1280,健康香蕉组织汁液无非特异性反应。用间接ELISA方法检测了采自广东、广西9个市(县)的香蕉产区的80个大田香蕉束顶病样本和25个荫棚香蕉试管苗样本,其中大田样本BBTV的带毒率为76.5%,试管苗样本的带毒率为8.0%。香蕉束顶病株不同部位BBTV含量检测表明,BBTV在病株中系统性分布,叶肉和中脉中的BBTV含量无差异。下部老叶与上部嫩叶相比,症状较轻,病毒含量也较低。
A indirect ELISA method for detecting banana bunchy top disease was developed by using the monoclonal antibody against the virus disease agent (BBTV). The leaf juice of diseased plants reacted postively with the McAb by this method when it was dilluted to 1:1280, but the leaf juice of healthy banana plants reacted negatively.
The samples of plants infected with BBTV in the field and the tube seedlings in vitro, collected from 9 cities or counties in Guangdong and Guangxi Provinces were detected by the indirect ELISA. The results showed that the samples from the tube seedlings and from field plants were infected by BBTV in rates of 8. 0% and 76. 5%, respectively.
The juice of different parts of banana plants infected with BBTV was detected with the McAb separately. The BBTV was systematically distributed the diseased plant and the concentration of BBTV in the midrids and blades was no different. The concentration of BBTV in older leaves with middle symptom was lower than that in the younger leaves with obvious symptom.
