摘要
目的 :探讨新生儿脐血中白细胞介素 - 2 (IL - 2 )、可溶性白细胞介素 - 2受体 (s IL - 2 R)、膜白细胞介素 - 2受体 (m IL - 2 R)、白细胞介素 - 6 (IL - 6 )和可溶性细胞间粘附分子 - 1(s ICAM- 1)的表达水平及其临床意义。方法 :以 EL ISA法检测新生儿脐血中 IL - 2、s IL - 2 R、m IL - 2 R、IL - 6和 s ICAM- 1含量 ,以 Ficoll- Hypaque常规分离产妇外周血单个核细胞 (PBMC)和脐血单个核细胞 (CBMC) ,用生物素 -链霉亲和素 (BSA)系统检测 m IL - 2 R表达水平。结果 :产妇外周血血清中 IL - 2、s IL - 2 R、m IL - 2 R、IL - 6和 s ICAM - 1含量分别为 (9.6 7± 4 .2 6 ) pg/ m l、(2 4 6 .5 0± 97.30 ) pg/ ml、 (3.73±1.0 3) %、(6 .6 7± 1.4 2 ) pg/ ml和 (5 6 0 .70± 136 .5 0 ) ng/ m l。新生儿脐血中 IL - 2、s IL - 2 R、m IL - 2 R、IL - 6和 s ICAM- 1含量分别为 (13.4 7± 3.4 0 ) pg/ ml、(15 5 .0 0± 83.5 0 ) pg/ m l、(1.30± 0 .4 5 ) %、(12 .87± 3.15 ) pg/ m l和 (32 1.2 0± 10 2 .2 0 ) ng/ ml。脐血 IL - 2、s IL - 2 R、m IL - 2 R、IL - 6、s ICAM- 1含量与产妇外周血相比 ,差异均有显著性 (P<0 .0 1)。结论 :脐血虽含有较丰富的天然造血细胞因子 。
Objective:To explore the expressive levels of interleukin-2 (IL-2),soluble interleukin-2 receptor (sIL-2R),membrane interleukin-2 receptor (mIL-2R),interleukin-6 (IL-6) and soluble intercellular adhesion molecule-1 (sICAN-1) in peripheral blood of parturient and cord blood of newborn.Methods:The levels of IL-2,sIL-2R,IL-6 and sICAN-1 in serum of peripheral blood of partrient and cord blood of newborn were all detected by ELISA.The peripheral blood clear cells (PBMC) of parturient and cord bloodmononuclear cells (CBMC) were isolated by routine Fioll-Hypaque,and the levels of mIL-2R on surface of PBMC and CBMC were detected by biotin-strptavidin (BSA).Results:The levels of IL-2,sIL-2R,mIL-2R,IL-6 and sICAM-1 in serum of peripheral blood of parturiet were (9.67±4.26)pg/ml,(246.50±97.30)pg/ml,(3.73±1.03)%,(6.67±1.42)pg/ml and (560.70±136.50)ng/ml.The levels of IL-2,sIL-2R,mIL-2R,IL-6 and sICAM-1 in serum of umbilical artery of newborn were (13.47±3.40)pg/ml,(155.00±83.50)pg/ml,(1.30±0.45)%,(12.87±3.15)pg/ml and(321.20±102.20)/ng/ml.The levels of IL-2,sIL-2R,IL-6 and sICAM-1 in serum of umbilical blood and mIL-2R on CBMC were all lower than those of parturient ( P <0.01).Conclusions:The almost active immunocytes in cord blood are infantile and undifferentiared,which surface sign and cytokines are infantile,and can be regar ded as one of effective,economical and safe origin of hematopoietic stem cells.The low powitive rate and light extent of graft versus host disease (GVHD) may be caused by these infantile and undifferentiated immunocytes.
