摘要
为检测新的凋亡相关基因PDCD5在狼疮性肾炎患儿肾组织中的表达 ,分析其与肾组织细胞凋亡的关系 ,探讨PDCD5在狼疮性肾炎发病中的作用 ,用免疫组化及图象分析的方法 ,检测19例狼疮性肾炎患儿肾组织及9例对照肾组织中PDCD5的表达 ;同时16/19例狼疮性肾炎和6/9例对照肾组织标本运用原位末端标记法(TUNEL)检测肾组织中的细胞凋亡。结果显示 :①在肾组织中可检测到PDCD5的表达 ,肾小管的表达量(0.20±0.02)强于肾小球的表达量 (0.09±0.01,P<0.05) ;②在狼疮性肾炎患儿肾组织中 ,PDCD5在肾小管的表达量 (0.13±0.01)强于肾小球的表达量 (0.07±0.01 ,P<0.05) ,PDCD5在肾小球的表达与对照组相比差异无显著性 ,在肾小管的表达明显低于对照组(P<0.05) ;③狼疮性肾炎患儿肾组织中肾小球、肾小管的凋亡细胞数均明显高于对照组(P<0.05)。本研究用免疫组化的方法首次检测了新的凋亡相关基因PDCD5在肾组织中的表达及分布 ;并发现在狼疮性肾炎患儿肾组织中PDCD5在肾小球的表达与对照组相比差异无显著性 ;在肾小管 ,虽然凋亡细胞数增加 ,但PDCD5的表达却下降。
s To detect the expression of a novel apoptosis-related gene PDCD5in the renal tissues of the children with lupus nephritis and analyze the correlation between PDCD5expression and apotosis of the renal tissues cells as well as investigate the role of PDCD5expression played in the pathogenesis of the paˉtients with lupus nephritis.The PDCD5expression in renal tissues was detected in19patients with lupus nephritis and9cases of the control group with immunohistochemical staining and pattern analysis.At the same time,the apoptosis occurred in the renal tissues was determined in16/19patients with lupus nephritis and6/9cases from the control group with in situ nick-end labeling techniques(TUNEL).The results showed as following(1)PDCD5expression could be detected in the renal tissues.The level of expression in renal tubule was much higher than that in glomeruli(0.20±0.02and0.09±0.01,respectively,P<0.05).(2)The level of PDCD5exprssion in renal tubule was also higher than that in glomeruli in the biopsy samples from patients with lupus nephritis(0.13±0.01and0.07±0.01,separately,P<0.05).There was no difference in PDCD5expression level in glomeruli between patients and control group.But the expression level in renal tubule of the patients was markedly lower than that in control group(P<0.05).(3)The number of apoptotic cells in the renal biopsy samples from patients with lupus nephritis(including in renal tubule and glomeruli)was significantly higher than that in control group(P<0.05).So the expression of the novel apoptosis-reˉlated gene PDCD5and its distribution in renal biopsy samples was initially assayed with immunohistochemical staining indicating that there was no difference of the expression level in glomeruli between patients and control group,but in renal tubule,the number of apoptosis cells was higher,with the PDCD5expression level significantly lower.
