摘要
采用SDS PAGE法对 16 2头二花脸母猪和 5 0头大约克母猪乳中一组高分子量蛋白质 (HMWP)进行了检测 ,计算了该位点基因型频率、等位基因频率和遗传多样性指数 ,并运用线性模型统计方法分析二花脸母猪该位点不同基因型与母猪泌乳性能的关系。结果表明 ,在猪种群中 ,乳中HMWP存在A (相对分子质量 14 6 80 0± 14 0 0 ) ,B ( 114 80 0± 4 0 0 ) ,C ( 10 86 0 0 )和D ( 10 14 0 0± 6 0 0 ) 4种带型 ,分别受 4个等位基因HA、HB、HC 和HD 控制。在二花脸猪乳中HMWP表现出BB、BD和DD 3种基因型 ,其频率分别为 0 6 6 6 7,0 2 716和 0 0 6 17;两个等位基因HB 和HD 频率分别为 0 80 2 5和0 1975 ;遗传多样性指数为 0 3170。大约克猪乳中HMWP表现出BB、AB和BC 3种基因型 ,其频率分别为 0 880 0 ,0 10 0 0和 0 0 2 0 0 ;等位基因HA、HB 和HC 的频率分别为 0 0 5 0 0 ,0 94 0 0和 0 0 10 0 ;遗传多样性指数为 0 1138。在二花脸猪种群中 ,不同HMWP基因型在乳糖浓度上存在显著差异 (P <0 0 5 ) ,但在乳蛋白浓度差异不显著 (P >0 0 5 )。
High molecular weight protein(HMWP)polymorphism of milk was investigated in 162 Erhualian sows and 50 Yorkshire sows by using SDS PAGE.The genotypic frequencies,allele frequencies and heterozygosities( h )in the locus of HMWP were calculated.The relationship between HMWP polymorphism and some lactation performance was analyzed with linear model in Erhualian sows.The results showed that four kinds of variant HMWP(A,B,C,and D bands)were observed to segregate among these sows.Apparent molecular weights of A,B,C and D were 146 800±1 400,114 800±400,108 600 and 101 400±600 respectively,controlled by allele H A,H B,H C and H D respectively.Three phenotypes(BB,DD and BD)were observed in Erhualian sows.The frequencies of genotype BB,BD and DD were 0 666 7,0 271 6 and 0 061 7 respectively in Erhualian sows,and the frequencies of allele H B and H D were 0 802 5 and 0 197 5 respectively.The frequencies of genotype BB,AB and BC,observed in Yorkshire sows,were 0 880 0,0 100 0 and 0 020 0 respectively,and the frequencies of allele H A,H B and H C were 0 050 0,0 940 0 and 0 010 0 respectively.The heterozygosities( h )of this locus in Erhualian and Yorkshire sows were 0 317 0 and 0 113 8 respectively.The effects of HMWP genotypes as a discrete variable were significant for lactose concentration( P <0 05),but not significant for milk protein concentration( P >0 05)in Erhualian sows.
