摘要
目的 研究乙酰胆碱对大鼠胃电节律的作用机制。方法 建立大鼠胃电节律失常模型 ,计算机采集并分析正常和模型大鼠胃电信号 ;肌间神经丛铺片 ,Kamovsky Root′s直接法显示肌间神经丛胆碱能神经 ,并经图像分析定量 ;放免法测定正常及模型组大鼠胃平滑肌组织中cAMP含量。结果 模型组肌间神经丛胆碱能神经分布面密度 (15 .2 1%± 5 .6 0 % )明显低于正常组 (2 4 .88%± 8.0 8% ) ,P <0 .0 1,而cAMP含量 (5 10 .4 9± 2 6 9.6 8)FM/mg明显高于正常组 (2 76 .6 5± 12 5 .97)FM/mg ,P <0 .0 1;胃慢波异常节律指数 (ARI) (18.0 1%± 11.0 8% )、频率变异系数 (CV) (36 .0 1%± 8.6 6 % )明显高于正常组 ,分别为 (4 .12 %±3.77% ) ,(2 1.5 5 %± 3.98% ) ,P <0 .0 1;且胆碱能神经含量与慢波异常节律指数和频率变异系数均呈显著负相关 (r=- 0 .90 9,P<0 .0 1与r =- 0 .6 4 2 ,P <0 .0 5 ) ,而cAMP含量与慢波异常节律指数和频率变异系数均呈显著正相关 (r=0 .90 2 ,P <0 .0 1与r=0 .733,P <0 .0 5 )。结论 模型组大鼠胃电节律失常与肌间神经丛胆碱能神经减少、cAMP增多有关 。
Objective To study the effects of acetycholine on electrogastric rhythmia of rats . Methods After rat models of electrogastric dysrhythmia were established, the gastroelectric slow waves were recorded and analyzed by computer. The levels of cAMP in the gastric smooth muscle of the normal group and the model group were measured by radioimmunoassary. Results The content of the cholinergic nerves in the model group(15.21±5.60)% was significantly reduced than that in the normal group( 24.88 ±8.08)%, P <0.01 . The level of cAMP in the model group(510.49±269.68)FM/mg was higher than that in the normal group (276.65±125.97)FM/mg, P <0.01. The ARI and CV in the model group were much higher than those in the normal group, P <0.01. The density of the cholinergic nerves were positively correlated to the ARI and CV in the model group, while the levels of cAMP were negatively correlated to the ARI and CV. Conclusions The gastroelectric dysrhythmia in the rat model is related to the reduction of cholinergic nerves, as well as the enhancement of cAMP. Acetylcholine decreases electric dysrythmia via cAMP pathway.
