摘要
目的研究肿瘤坏死因子(TNF)对体外培养的猪主动脉内皮细胞(PAEC)与单核细胞(MC)黏附的影响及几丁聚糖的拮抗作用。方法通过测定MC髓过氧化物酶活性的方法,对黏附于PAEC单层上的MC进行定量。结果TNF在5~500U/ml时呈剂量依赖性地诱导PAEC与MC的黏附,TNF浓度为250U/ml时促黏附达最大值(81.98%)。TNF与内皮细胞共育1h,即可明显促进黏附作用(66.8 %),4h时达最大刺激效应(77.5 %)。几丁聚糖在浓度为20~2000μg/ml时,呈剂量依赖性地拮抗TNF诱导的黏附作用,在2000μg/ml时抑制率达22.99 %。结论几丁聚糖具有抑制由TNF诱导的PAEC与MC黏附的作用。
Objective To study inhibitory effects of chitosan on tumor necrosis factor(TNF)induced adhesion reaction of cultured pig aortic endothelial cells(PAEC)with monocytes(MC)in vitro.Methods The assay was based on the measurement of the myeloperoxidase activity in monocytes that had adhered to PAEC layers.Results With concentration from5to500U/ml,TNF was shown to induce adhesion reaction of cultured PAEC with MC in a dose-dependent manner.The effect of TNF reached maximum(81.98%)at250U/ml.After1h incubation of PAEC,addition of TNF significantly stimulated the adhesion reaction of RAEC with MC(66.8%).After2h incubation the effect reached maximum(77.5%),then the effect slightly decreased.With concentration from20to2000μg/ml,chitosan was found to antagonize induced adhesion reaction.Its maximal rate of inhibition was22.99%at2000μg/ml.Conclusions Chitosan may decrease the TNF induced adhesion reaction of PAEC with MC.
