摘要
HCAP1基因系人类染色体 17p13 .3区带克隆的新肝癌相关基因。已发现人类多种肿瘤存在此区带的杂合性缺失。本研究应用脂质体介导法将HCAP1基因转染T淋巴瘤Jurkat细胞系 ,经G418筛选 ,获得稳定表达外源HCAP1基因的细胞。用台盼蓝拒染试验计数活细胞 ,绘制生长曲线 ,进行软琼脂集落形成试验。结果显示 :与转染空载体质粒 pBK/CMV的细胞比较 ,转染外源HCAP1基因的Jurkat细胞增殖速率明显减慢 ,细胞倍增时间延长 ,在软琼脂上的集落形成能力下降 (P <0 .0 1)。结论 :外源HCAP1基因产物对Jurkat细胞的增殖有明显的抑制作用。
HCAP1 is a novel hepatic cancer related gene lo ca ted on human chromosome 17p13.3. The loss of heterozygosity occurred at 17p13.3 in various human cancers. In order to investigate the effects of exogenous HCAP1 gene products on cell proliferation of T lymphoma Jurkat cell line, HCAP1 gene was transfected into Jurkat cells mediated by liposome, and the cells stably exp ressing exogenous HCAP1 were screened with G418. The effects of HCAP1 products o n cell proliferation were assessed by viable cell count, cell growth curve and c olony formation assay in soft agar. The results showed that the HCAP1 transgenic Jurkat cells displayed slow growth rate, extended doubling time and reduced col ony formation capability, as compared with the cells transfected with pBK/CMV em pty vector (P<0.01). It is concluded that exogenous HCAP1 gene products coul d inhibit the proliferation of Jurkat cells.
