常见消化系统肿瘤CD177及相关基因表达与意义的生物信息学分析 被引量:2

Expression of CD177 and related genes in common gastrointestinal cancers and its clinical significance: a bioinformatic analysis

摘要 目的探讨嗜中性粒细胞抗原CD177在常见消化系统恶性肿瘤(食管癌、结肠癌和胰腺癌)中的表达情况与患者预后的关系,并分析与其相关的基因和细胞通路。方法自公共数据库Gene Expression Omnibus(GEO)及The Cancer Genome Atlas(TCGA)中提取相应癌症数据集,以获取患者信息及mRNA表达数据,借助R2生物信息分析平台,以Kaplan-Meier法绘制生存曲线,分析CD177表达情况与患者预后的关系,筛选出与CD177表达显著相关的基因并进行Kyoto Encyclopedia of Genes and Genomes(KEGG)通路分析。结果在3种癌症数据集中(食管癌75例,结肠癌174例,胰腺癌132例),CD177的mRNA高表达患者的总体生存率均低于低表达患者(P=0.050,P=0.045,P=0.018)。此外,与CD177表达相关的基因在3个数据集中分别有11 927、4 100和7 493个。KEGG通路分析发现,CD177高表达时除正常消化功能的信号通路被激活外,还涉及多种炎性反应机制相关的信号通路。结论 CD177的表达水平与食管癌、结肠癌和胰腺癌患者的总体生存率相关,可能成为研究患者预后的潜在分子标志物。 Objective To investigate the expression of neutrophil antigen CD177 in common digestive malignancies(esophageal cancer,colon cancer and pancreatic cancer)and its clinical significance and to analyze the correlated genes and cellular pathways with bioinformatics.Methods The corresponding cancer datasets were extracted from the public database Gene Expression Omnibus(GEO)and The Cancer Genome Atlas(TCGA),including patient information and the mRNA expression data.The Kaplan-Meier survival curve was plotted using the R2 bioinformatics analysis platform,and the relationship between CD177 expression and patient’s prognosis was analyzed.Genes significantly associated with CD177 expression were selected and the Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis was performed.Results In the datasets of three cancer types(75 cases of esophageal cancer,174 cases of colon cancer,and 132 cases of pancreatic cancer),the overall survival rates of patients with high CD117 mRNA expression were significantly lower than those with low CD117 expression(P=0.050,P=0.045,P=0.018).In addition,there were 11 927,4 100,and 7 493 genes significantly associated with CD177 expression in the three datasets,respectively.Moreover,analysis of the KEGG pathway revealed that in addition to the over-activation of signaling pathways related to normal digestive function,multiple signaling pathways involved in inflammatory response mechanisms were also significantly related to high CD177 expression.Conclusion The expression level of CD177 is correlated with the overall survival rate of patients with esophageal cancer,colon cancer or pancreatic cancer,which suggests that CD117 may be a potential molecular marker for patients′prognosis.
