摘要
目的探讨不同浓度脂多糖(lipopolysaccharide,LPS)对小鼠血液细胞因子水平及小肠组织形态的影响。方法将雄性ICR小鼠随机分为5组,对照组经腹腔注射生理盐水,4个试验组分别经腹腔注射2.5、5、10、20 mg/kg LPS,注射6 h后,经小鼠眼球采血,分离血清,ELISA法检测小鼠血清中炎性细胞因子TNF-α、IL-1β、IL-4、IL-6和IL-8水平;同时采集小鼠十二指肠、空肠、回肠和盲肠组织样品,做组织切片后进行HE染色,测量小肠段绒毛高度(villus height,VH)和隐窝深度(crypt depth,CD)。结果试验组小鼠肠道碱性磷酸酶(intestinal alkaline phosphatase,IAP)及细胞因子水平在LPS浓度为5 mg/kg时达到高峰,随着LPS浓度的升高,细胞因子分泌受到抑制;与对照组相比,十二指肠、空肠和回肠组织VH值降低、VH/CD比值下降。结论小鼠在注射5 mg/kg LPS时,可诱导肠黏膜损伤,使得各细胞因子浓度达到最高,后续可作为炎症性肠炎模型适宜的诱导剂量。
Objective To investigate the effect of lipopolysaccharide(LPS)at various concentrations on the serum cytokine level and morphology of intestinal tissue of mice.Methods Male ICR mice were randomly divided into five groups.The mice in control group were injected i.p.with physiological saline,while those in four test groups with LPS at concentrations of 2.5,5,10 and 20 mg/kg respectively.Serum samples were collected 6 h after injection and determined for TNF-α,IL-1β,IL-4,IL-6 and IL-8 levels by ELISA.Specimens of intestinal wall of the duodenum,jejunum,ileum and cecum were prepared for histological examination with HE staining,and the VH(villus height)and CD(crypt depth)were measured.Results The intestinal alkaline phosphatase(IAP)and cytokine levels reached the maximums at a LPS concentration of 5 mg/kg LPS,with no further increase with the increasing LPS concentration.Compared with those in control group,the VH and VH/CD values of duodenum,jejunum and ileum decreased.Conclusion LPS at a concentration of 5 mg/kg induced intestinal mucosal injury in mice and increased the concentrations of various cytokines to the maximums,which might be used as an appropriate dosage for induction of inflammatory enteritis in animal.
