摘要
利用响应面设计试验优化灵芝孢子多糖的热水浸提工艺条件。以超临界CO_2萃取过灵芝孢子油后的灵芝孢子粉为原料,采用单因素试验,探究提取温度、提取时间、液料比对灵芝孢子多糖提取率的影响。在单因素试验的基础上,以多糖提取率为响应值,利用Box-Behnken试验设计和响应面分析法对提取条件进行优化,以确定最佳的提取工艺:提取温度91℃,提取时间3.3 h,液料比15.8︰1 (mL·g^(-1))。在此条件下多糖的提取率达到3.69%,与理论值相对误差0.33%。结果表明利用响应面法优化灵芝孢子多糖的热水浸提工艺是可行的。
Using response surface methodology to optimize hot water extraction conditions of polysaccharidies from Ganoderma Lucidum spores,based on Ganoderma lucidum spores which had been extracted Ganoderma Lucidum spores oil with supercritical carbon dioxide,using single factor experiment to investigated the effect of extraction temperature,extraction time and liquid-to-solid ratio on the yield of polysaccharides.Based on the single factor experiment,using the yield of polysaccharides as the response value,the extraction conditions by Box-Behnken design and response surface methodology to determine the optimal extraction conditions are optimized.The result show that the optimal extraction conditions are extraction temperature 91℃,extraction time 3.3 h and the ratio of liquid-to-solid 15.8︰1(mL·g-1).The yield of polysaccharides is 3.69%under this conditions,compared with the theoretical value,the relative error is 0.33%.The results indicate that it is reliable,using response surface methodology to optimize hot water extraction conditions of polysaccharidies from Ganoderma lucidum spores.
