摘要
目的探讨香青兰总黄酮对低氧环境所致大鼠肺损伤的保护作用。方法将60只SD大鼠随机分为正常组(CG)、高原组(MG)、阳性组(NE)、香青兰总黄酮高剂量组(DML.H)、香青兰总黄酮中剂量组(DML.M)、香青兰总黄酮低剂量组(DML.L)。除正常组(10只)外,其余50只大鼠建立慢性高原病模型。各组进行相应干预。测定各组大鼠肺动脉压力,血清中EPO、VEGF及肺组织SOD、MDA和GSH-Px活性,观察大鼠肺组织病理学改变。结果与正常组比较,慢性高原病大鼠肺动脉压力升高,血清EPO、VEGF含量上升,肺组织中SOD、GSH-Px含量降低,MDA含量升高(P<0.05)。与高原组比较,香青兰总黄酮各剂量组大鼠肺动脉压力不同程度下降(P<0.05),血清中EPO、VEGF含量降低(P<0.05),肺组织SOD、GSH-Px含量升高(P<0.05),MDA含量降低(P<0.05)。结论香青兰总黄酮可改善由缺氧引起的肺动脉高压,减少氧化应激,增强抗氧化机制,对低氧环境所致肺损伤具有一定的保护作用。
Objective To explore the protective effect of total flavonoids of Dracocephalum Moldavica L.on hypoxia-induced lung injury in rats.Methods 60 SD rats were randomly assigned into the following groups:control group,model group,nifedipine-treated group(NE),and high-dose group(DML.H),medium-dose group(DML.M)and low-dose group(DML.L)of total flavonoids of Dracocephalum Moldavica L.Excluding the control group(10 rats),the model of chronic mountain sickness was established with the other 50 rats.Each group of rats received corresponding intervention.Pulmonary artery pressure,the content of EPO and VEGF in serum,and the content of SOD,MDA and GSH-Px in lung tissues were determined and the pathological changes in lung tissues of rats were observed in each group.Results Compared with the control group,the pulmonary artery pressure increased,the content of EPO and VEGF in serum increased,the content SOD and GSH-Px in lung tissues decreased,and the content of MDA increased in rats with chronic mountain sickness(P<0.05).Compared with the model group,the pulmonary artery pressure decreased to varying degrees(P<0.05),the content of EPO and VEGF in serum decreased(P<0.05),the content of SOD and GSH-Px in lung tissues increased(P<0.05),and the content of MDA decreased(P<0.05)in the DML.H,DML.M and DML.L groups.Conclusion The total flavonoids of Dracocephalum Moldavica L.improves pulmonary arterial hypertension caused by hypoxia,reduces oxidative stress and enhances the antioxidant mechanism.It has certain protective effect on lung injury caused by hypoxic environment.
