黄芪总皂苷对CCl_4致小鼠急性肝损伤的保护作用 被引量:2

摘要 目的:探讨黄芪总皂苷对四氯化碳(CCl_4)诱导的小鼠急性肝损伤的保护作用。方法:将60只小鼠随机分为正常组、模型组、联苯双酯组(150 mg/kg),黄芪总皂苷高、中、低剂量组(200 mg/kg、100 mg/kg、50 mg/kg),每组10只,正常组和模型组给予等量溶媒,每天1次,连续10 d。末次给药2 h后,每只小鼠腹腔注射10 mL/kg以橄榄油配制的5%CCl_4溶液。16~18 h后,自小鼠后眼眶静脉丛采血,检测血清中超氧化物歧化酶(SOD)、丙二醛(MDA)含量,计算肝脏及脾脏指数,HE染色观察肝脏组织病理形态学的改变,免疫组化法观察肝脏细胞中IL-1和TNF-α的表达水平。结果:与正常组相比,模型组小鼠血清MDA明显升高,SOD明显降低;与模型组比较,黄芪总皂苷各组MDA显著降低,SOD显著升高,肝脏组织形态结构改变明显减轻,肝脏IL-1和TNF-α的表达被显著抑制。结论:黄芪总皂苷对CCl_4致小鼠急性肝损伤有显著保护作用,其机制可能与清除自由基、减轻肝细胞凋亡和抗氧化损伤有关。 Objective:To investigate the protective effects of astragalus saponins on acute liver injury induced by carbon tetrachloride(CCl4)in mice. Methods:60 mice were randomly divided into normal group,model group,biphenyl diester group(150 mg/kg),astragalus total saponin high,medium and low dose group(200 mg/kg,100 mg/kg,50 mg/kg),10 mice in each group. The normal group and the model group were given the same amount of solvent,1 time a day for 10 days. 2 h after the last administration each mouse was intraperitoneally injected with 10 mL/kg of 5% CCl4 solution prepared with olive oil. After 16 ~18 h,blood was collected from posterior orbital venous plexus of mice,then the contents of superoxide dismutase(SOD)and malondialdehyde(MDA)in serum were detected. Indice of liver and spleen were calculated and HE staining was used to observe the pathological changes of liver tissue. Immunohistochemistry was used to detect the expressions of IL-1 and TNF-α in liver tissue. Results:Compared with the normal group,MDA content was decreased,and SOD was significantly increased in model group. Compared with the model group,MDA content and the expressions of IL-1 and TNF-α were significantly decreased while SOD content was significantly increased in astragalus saponins groups. Morphological and structural changes of liver tissue in astragalus saponins groups were significantly better than those in model group.Conclusion:Astragalus saponins has significant protective effects on acute liver injury induced by CCl4,which may be related to scavenging free radicals,reducing hepatocyte apoptosis and anti-oxidative injury.
