摘要
目的观察丹参提取物对放射性肺损伤大鼠的预防及保护作用,探讨其可能的作用机制。方法 72只健康雌性SD大鼠随机分为3组:空白对照组(A组)、单纯照射组(B组)、照射加丹参提取物组(C组)。开始照射前1周,A组和B组灌胃生理盐水,C组灌胃丹参提取物,至照射2周后结束给药,共灌胃3周。给药1周后,B、C 2组大鼠经直线加速器以12 Gy单次照射,分别于照射后第30、60、120天,随机处死6只大鼠,检测比较各组大鼠一般情况,ELISA检测血清SOD含量,硫代巴比妥酸化学比色法测定MDA含量及右肺系数。结果 B组大鼠照射后出现精神萎靡、食欲下降、对刺激的反应性下降等症状,且随时间的延长,症状不断加重;C组大鼠较B组精神状态有明显改善;照射后第30、60、120天,C组大鼠血清SOD水平接近于A组,显著高于B组(P<0.05);照射后第30、60天,C组大鼠右肺系数显著低于B组,说明丹参提取物组可阻止放射性大鼠肺系数的增长;照射后第30、60、120天,C组大鼠MDA含量显著低于B组(P<0.05)。结论丹参提取物可明显改善放射性肺损伤大鼠的生活精神症状,提高大鼠血清SOD水平,降低MDA含量和右肺系数,从而起到保护作用。
Objective To observe the effect of Salvia Miltrorrhiza extracts of preventing radiation-induced lung injury in rats and explore its feasibility and mechanism as a radiation-induced lung injury prevention measure. Methods 72 healthy adult female Wistar rats were randomly divided into three groups; control group( A),the irradiation group( B),irradiation and Salvia Miltrorrhiza extracts group( C). One week before irradiation,A and B group were treated with physiological saline by gavage,C group were treated with Chinese medicine daily by gavage. The drugs were stopped after two weeks 12 Gy irradiation,and length of medication use was three weeks. One week after administration,B and C group were treated with 12 Gy irradiation by linear accelerator.,six rats were randomly selected and killed after 30 days,60 days,120 days irradiation,and the general conditions of rats were detected and compared. SOD content was detected by ELISA,MDA content and the right lung coefficient were detected by thiobarbituric acid colorimetric method. Results The rats in B group after irradiation appeared such symptoms as listlessness,loss of appetite,decreased response to stimulation,and the symptoms got worse with the extension of time; the mental state of rats in C group had more improvement than those in B group; after 30 days,60 days,120 days irradiation,the SOD levels of C group was close to A group,and significantly higher than B group with significant difference( P < 0. 01);after 30 days,60 days irradiation,the right lung coefficient of rats in C group was significantly lower than B group( P < 0. 05),which showed that Salvia Miltrorrhiza extracts could prevent the growth of rat lung coefficient; after 30 days,60 days,120 days irradiation,the MDA content of rats in C group was higher than that in B group,and there was significant difference( P < 0. 05).
