摘要
目的:观察不同剂量射线对大鼠肺泡巨噬细胞增殖能力的影响,为放射性肺损伤细胞模型建立奠定前期基础。方法:使用高能直线加速器X射线以不同剂量射线(0 Gy、2 Gy、4 Gy、6 Gy)照射大鼠肺泡巨噬细胞,分别于照射后24 h、48 h、72 h后观察细胞镜下形态,并用MTT法检测细胞增殖能力。结果:在不同时间节点,一个视野内,照射组细胞数量均少于正常组(0 Gy组),从细胞形态来看,0 Gy组细胞表现为大鼠肺泡巨噬细胞呈串样生长的特点,而照射组细胞则呈单个分散样生长;经MTT法检测,照射后各时间节点测定OD值分别为:0 Gy组,0.139、0.165、0.205; 2 Gy组,0.112、0.127、0.151; 4 Gy组,0.109、0.121、0.138; 6 Gy组,0.088、0.094、0.099。其中不同剂量射线照射组与正常组相比,吸光度值均有差异,P=0.000 <0.05;不同时间点,24 h,48 h,72 h,各组吸光度之间均有差异,P=0.000 <0.05;不同剂量照射组之间两两相比,2 Gy组与4 Gy组差异不明显,P=0.052> 0.05,6 Gy组与其他两组之间差异显著。结论:射线会对大鼠肺泡巨噬细胞的生长特性和增殖能力产生影响,使其串样生长特性消失,其中6 Gy剂量射线对细胞所产生的影响最大。
Objective:To observe the effect of different doses of radiation on the proliferation of the cells,and to lay a foundation for the establishment of the radiation lung injury cell model. Methods:The alveolar macrophages of rats were irradiated with different doses of radiation (0 Gy,2 Gy,4 Gy,6 Gy) by high energy linear accelerator (X ray).The morphology of the cells under the cell microscope was observed after 24 h,48 h and 72 h after irradiation,and the cell proliferation ability was detected by MTT method. Results:The number of cells in the irradiated group was less than that in the normal group (group 0 Gy) at the different time nodes.In the form of cell morphology,the cells in the 0 Gy group showed a series of growth characteristics of the alveolar macrophages in the rats,while the cells in the irradiated group showed a single dispersed growth.After the MTT test,the measured values of each time node in 0 Gy were 0.139,0.165,0.205 and 0.112,0.127,0.151 in 2 Gy and 0.109,0.121,0.138 in 4 Gy,and 0.088,0.094,0.099 in 6 Gy.Compared with the normal group,the absorbance values of different doses were different, P =0.000<0.05.In 24 h,48 h,72 h,the absorbance of each group were different, P =0.000<0.05.The difference between group 2 Gy and 4 Gy group was not obvious, P =0.052>0.05.There was a significant difference between the groups in 6 Gy group and other two. Conclusion:The radiation can affect the growth and proliferation of alveolar macrophages in rats,and disappears the characteristics of the growth of the rat alveolar macrophages,in which the 6 Gy dose rays have the greatest impact on the cells.
