地黄饮子加减方对D-半乳糖致衰老模型小鼠抗氧化作用的影响 被引量:6

摘要 目的:观察地黄饮子加减方对D-半乳糖致衰老模型小鼠抗氧化能力的影响,探讨地黄饮子加减方的抗衰老作用。方法:①采用7月龄ICR雄性小鼠40只为研究对象,采用D-半乳糖腹腔注射制备亚急性衰老小鼠模型,随机分为模型组、模型VE组、模型地黄组,每组保证10只,并设置正常组10只为对照。②利用跳台实验方法检测各组小鼠的学习、记忆能力。③采用HE染色观察各组小鼠脑组织形态改变。④检测各组小鼠血清中超氧化物歧化酶(SOD)、乙酰胆碱酯酶(AchE)、谷胱甘肽过氧化物酶(GSH-Px)指标数值。结果:①模型组经地黄饮子加减方干预后,学习能力测试,小鼠跳台实验的错误次数减少(P<0.01);记忆能力测试,小鼠跳台的错误次数减少(P<0.01)。②与模型组比较,地黄饮子加减方能够有效缓解小鼠脑组织病理损伤状况。③与模型组比较,地黄饮子加减方能够使小鼠血清中SOD及GSH-Px含量显著提高(P<0.01),使血清中AchE的含量明显降低(P<0.01)。结论:地黄饮子加减方能够改善模型小鼠的学习记忆能力,减轻模型小鼠脑组织神经元脱失状况,提高抗氧化能力,从而起到减缓衰老的作用。 Objective:To observe the effect of Modified Dihuang Yinzi Decoction on the antioxidant capacity of D-galactose(D-gal)-induced aging mice,and to explore the effect of Modified Dihuang Yinzi Decoction on anti-aging.Methods:①Seven-month-old ICR male mice were used as the study subjects.The subacute aging mouse model was prepared by intraperitoneal injection of D-galactose and all the mice were randomly divided into model group,model VE group and model Dihuang group.Ten mice in each group were guaranteed and 10 mice in normal group were set as the control.②Platform test method was used to test the learning and memory ability of mice in each group.③The brain tissue morphology of each group was observed by HE staining.④The values of superoxide dismutase(SOD),acetylcholinesterase(AchE)and glutathione peroxidase(GSH-Px)in the serum of each group were detected.Results:①After the intervention of Modified Dihuang Yinzi Decoction,the number of errors in the mouse platform test the learning ability test was reduced(P<0.01).In the memory ability test,the number of errors in the mouse platform was reduced(P<0.01).②Compared with the model group,Modified Dihuang Yinzi Decoction can reduce the pathological damage of brain tissue in mice.③Compared with the model group,Modified Dihuang Yinzi Decoction can significantly increase the content of SOD and GSH-Px in the serum of mice(P<0.01),and significantly reduce the content of AchE in serum(P<0.01).Conclusion:Modified Dihuang Yinzi Decoction can improve the learning and memory ability of model mice,reduce the neuronal loss of brain tissue of model mice,improve the antioxidant capacity and ultimately slow down the aging process.
