人甲状腺未分化癌细胞裸鼠皮下移植模型的建立 被引量:2

摘要 目的 :建立人甲状腺癌细胞裸鼠皮下移植模型。方法 :用人甲状腺未分化癌细胞系 TA- K细胞悬液裸鼠皮下注射及瘤块皮下植入的方法建立人甲状腺未分化癌裸鼠皮下移植模型 ,观察移植后肿瘤生长特点及组织学特征。结果 :皮下注射法与肿瘤块植入法均可出现皮下肿瘤 ,染色体检查证实裸鼠皮下移植瘤的染色体组型与人体肿瘤一致。皮下注射方法肿瘤注射后经过 1 1~ 1 8d的潜伏期后才进入倍增期 ,瘤块皮下植入方法则跨过潜伏期直接进入倍增期。结论 :利用人甲状腺未分化癌细胞系 TA- K成功建立人甲状腺癌裸鼠皮下移植模型 ,肿瘤块植入法比皮下注射法省时、经济、稳定 。 Objective:To describe the development of an animal model by using ectopic implantation of human anaplastic thyroid cancer in nude mice. Methods:Human anaplastic thyroid cancer cell line TA K was injected in the subcuitis in 6 nude mice. Tumor were found grown 11 18days later. Tumor tissue were acquired and transplanted in subcuitis in 4 nude mice. Mice were observed carefully and sacrificed when found moribund, and autopsy and histology were performed subsequently. Results:The implantation of human anaplastic thyroid cancer cells in the subcuitis permit expression 100% tumor grown. Tumor tissue transplantation group did not pass through latent phase, whereas tumor cell injection group did. Histological examination confirmed that the implanted tumor chromosome was identical to human. Conclusions:An animal model was established when human anaplastic thyroid cancer cell line TA K was implanted into nude mice. Tumor tissue transplantation could make the establishment of model more fast, more stable and more thrifty.
