摘要
目的:探讨甲状腺癌细胞中BCORL1对E-cadherin的调控及其对侵袭转移能力的影响。方法:采用Western Blot技术检测人甲状腺癌细胞TA-K、TPC-1和FTC-133中BCORL1及E-cadherin的表达;应用siRNA特异性干扰TA-K细胞中BCORL1的表达,Western Blot和qRT-PCR技术检测BCROL1和E-cadherin表达变化;Transwall侵袭和迁移小室检测BCORL1对TA-K细胞侵袭转移能力的影响。结果:BCORL1在TA-K细胞中的表达水平明显高于TPC-1和FTC-133,而E-cadherin表达水平则明显低于TPC-1和FTC-133;特异siRNA能显著下调BCORL1mRNA(t=14.025,P<0.000 1)和蛋白表达,导致E-cadherin mRNA(t=8.004,P<0.000 1)和蛋白表达升高;下调TA-K细胞中BCORL1表达能显著减弱细胞侵袭和迁移能力(t=5.942,P=0.000 3;t=7.115,P=0.000 1)。结论:人甲状腺癌细胞中BCORL1可能通过转录抑制调控E-cadherin表达,从而促进肿瘤侵袭转移。
Objective:To investigate the regulation of E-cadherin by BCORL1 and its effect on invasion and metastasis of thyroid cancer cells.Methods:We tested the expression of BCORL1 and E-cadherin by using Western blot in TA-K,TPC-1and FTC-133 thyroid cancer cell lines.TA-K cells were transfected with BCORL1 siRNA,Western blot and qRT-PCR were used to examine the change of BCORL1 and E-cadherin expression.Transwell invasion and migration assays were used to test the effect of BCORL1 on TA-K cell invasion and metastasis abilities.Results:The expression of BCORL1 was higher in TA-K cells than those in TPC-1and FTC-133 cells,but expression of E-cadherin was lower in TA-K cells than TPC-1and FTC-133 cells.siRNA significantly decreased the expression of BCORL1mRNA(t=14.025,P〈0.0001)and protein level,and increased exrepssion of E-cadherin mRNA(t=8.004,P 0.0001)and protein level.BCORL1 knockdown obviously reduced the number of invasion and migration cells(t=5.942,P=0.000 3;t=7.115,P=0.000 1,respectively).Conclusion:BCORL1promotes invasion and metastasis of thyroid cancer cells by down regulation of E-cadherin through transcriptional re-pression.
