摘要
建立了一种检测鱼组织中氟甲喹对映体的高效液相色谱方法。考察了流动相组成、流动相中乙醇比例、三乙胺含量、柱温、流速等条件对氟甲喹对映体分离的影响。确定最佳检测条件为:含有0.3%三乙胺的乙醇为流动相,流速0.5 mL/min,柱温20℃,检测波长254 nm,在此条件下,氟甲喹对映体分离度为2.64,氟甲喹对映体添加回收率均大于81.80%,相当标准偏差均小于6.38%。S-(-)-FMQ和R-(+)-FMQ的检出限分别为0.05μg/g和0.04μg/g,定量限分别为为0.08μg/g和0.07μg/g。该方法适用于鱼组织中氟甲喹对映体的快速检测。
In this paper, an enantioselective determination method was established for the flumequine in fish tissues by using high performance liquid chromatography. The effects on the detection method by mobile phase composition, the ratio of ethanol in mobile phase, triethylamine content, column temperature and flow rate were investigated. The optimized chromatographic conditions were that the mobile phase was ethanol containing 0.3%triethylamine with a flow rate of 0.5 mL/min and detection at the wavelength of 254 nm, the column temperature was set at 20 ℃. The separation degree of flumequine enantiomers was 2.64 under the optimized conditions. The average recoveries of flumequine enantiomers were higher than 81.80%and the RSDs were lower than 6.38. The LODs of S-(-)-FMQ and R-(+)-FMQ were 0.05μg/g and 0.04μg/g, the LOQs of S-(-)-FMQ and R-(+)-FMQ were 0.08μg/g and 0.07μg/g, respectively. This method showed high sensitivity, good recovery rate and simple pretreatment, it was applicable for the rapid detection of flumequine enantiomers in fish tissues.
