狗牙周膜细胞三维立体培养的体外实验研究 被引量:12

摘要 目的 :探讨异体脱矿松质骨基质 (CBM )和纳米羟基磷灰石材料 (nHAC)两种支架材料应用于牙周组织工程的可行性 ,为建立牙周组织工程的动物模型及下一步进行牙周组织工程的动物实验奠定实验基础。方法 :组织块法培养狗牙周膜细胞 ,传代扩增后 ,接种于CBM和nHAC 2种三维支架上 ,体外继续培养 3d ,进行细胞计数和扫描电镜观察。结果 :狗牙周膜细胞在 2种支架材料上均能形成良好贴附并增殖 ,扫描电镜可见 2种支架材料均具有良好的多孔网状结构 ,细胞在支架材料上生长旺盛 ,伸展充分。结论 Objective:To evaluate the feasibility of two materials used as scaffolds in periodontal tissue engineering. Methods: Dog periodontal ligament cells cultured in vitro were collected and seeded on three-dimensional framework of cancellous bone matrix (CBM) or nano-HAp/collagen (nHAC) . The cell growth in the scaffolds was observed by cell counting and scanning electronic microscope. Results: Porous structure of the two materials and adhesion and eugonic growth of cells in the scaffolds were observed by scanning electronic microscope.The cell number was doubled in both scaffolds in 72 h culture. Conclusion: It may be feasible to use CBM or nHAC as the scaffolds for periodontal tissue engineering.
