大鼠不同年龄及再生肝的肝细胞核倍体分析

摘要 本文利用细胞分光光度技术定量测定细胞学涂片中单个肝细胞DNA含量的方法,观察了不同年龄正常大鼠及大鼠部分肝切除后再生肝的肝细胞DNA含量情况。实验表明正常大鼠出生后,随着年龄的增长肝细胞核倍体逐渐从二倍体向四倍体发展;行2/3肝叶切除术后48小时,二倍体肝细胞和双核细胞明显减少,四倍体细胞增多,显示在增生刺激下促进了肝细胞多倍体化过程。最后,对肝细胞多倍体化的形成及其生物学意义进行了讨论。 The suspension of rat intact liver cells at different age or with liver regeneration was prepared. The isolated hepatocytes were smeared. The number of nuclei per cell was determined and the DNA content of each single nucleus was measured by a microspectrophotometer at a wavelength of 560nm. The transformation of the diploid population of hepatocytes in newborn animals into a practically exclusive polyploid took place during their postnatal growth. Analysis of rat hepatocytes undergoing liver regeneration following two-thirds partial hepatectomy revealed that there was an increase in tetraploid cells and a decrease in diploid and binuclear ones. The polyploidization appeared to be temporarily accelerated under the stimulus of proliferation.
