姜黄素诱导的平滑肌细胞凋亡在球囊损伤后内膜增生中的作用 被引量:6

Effects of Apoptosis of Vascular Smooth Muscle Cells Induced by Curcumin on Intima Hyperplasia after Injuring by Balloon

摘要 目的:观察姜黄素对家兔髂动脉球囊损伤后血管内膜增生的影响。方法:新西兰大白兔30只,随机分为3组:对照(C1)组、阿托伐他汀(G2)组、姜黄素(G3)组,每组各10只。所有实验组在高脂饮食基础上均予灌胃给药,其中G3组予姜黄素100mg/(kg·d),G2组予阿托伐他汀2.5mg/(kg·d),G1组予生理盐水5ml/d。1周后制作髂动脉球囊损伤模型,5周后取目的血管,经苏木精-伊红、弹力纤维染色观察内膜增生情况,同时电镜观察细胞凋亡,并检测凋亡基因p53、Caspase3。结果:病理切片图像分析显示内膜增生程度由强到弱为:G1组>G2组>G3组。在G3组p53表达明显增高,其他两组无差异(P>0.05)。Caspase3表达以G3组最高。电镜观察G3组平滑肌细胞可呈早中期凋亡现象。结论:姜黄素具有诱导血管平滑肌细胞凋亡作用,在一定程度上抑制血管内膜增生。 Objective:To investigate the effects of curcumin on intima hyperplasia of rabbit's iliac artery after injuring by balloon. Methods:Thirty New Zealand rabbits were divided randomly into 3 groups: control group (G1), atorvastatin group (G2) and curcumin group (G3). Each group had 10 rabbits. Rabbits in G1,G2,G3 were intragastrically perfused by normal saline, atovarstain 2.5 mg/(kg·d) and crucumin 100 mg/(kg·d) respectively. Model of iliacartery injury was established after gastric perfusing one week.On the 5th week,the target vessel was taken out.The intima hyperplasia was observed by microscopy through HE and special elastic fibers staining.Cell apoptosis was determined with electron microscopy, and apoptosis Gene P53, Caspase 3 were detected. Results:The area of neointima was the highest in G1, second G2 and the lowest G3. The expression of gene P53 in G3 was significantly abundant (P<0.01). There was no difference (P>0.05) between the other two groups.The expression of gene Caspase3 in G3 was the most abundant. The apoptosis of vascular smooth muscle cell (VSMC) in G3 was observed on the prophase and metaphase by electron microscopy. Conclusion: Curcumin may induce apoptosis of VSMC and suppress the proliferation of intima significantly.
