摘要
目的 观察ALT连续监测法不同试剂盒的可比性 ,规范酶活性浓度测定。方法 选用四种方法学相同的试剂盒同时在自动生化分析仪上测定后进行比较。结果 台湾元生、北京中生和上海长征公司试剂盒之间在 <80 0U/L内无显著性差异 (P >0 0 5 ) ,而标佳产品有差异 ,但无非常显著性差异 (0 0 1 <P <0 0 5 )。当ALT >80 0U/L时 ,各试剂盒间均有极显著性差异 (P <0 0 1 )。高活力血清标本经稀释后测定 ,各试剂盒间无显著性差异(P >0 0 5 )。结论 酶活力测定时 ,选择适宜试剂盒 ,根据酶反应线性期的反应速度计算酶活性浓度 ,观察试剂空白吸光度、动力学空白。
Objective To compare different ALT diagnosis kit for the determination of enzyme activity. Methods 4 different ALT diagnosis kits using the same detection methods were chosen for comparison in a automatic biochemical analyzer.Results In Taiwan Unison and Beijing Zhong Sheng and Shanghai Long March ALT diagnosis kits showed no significant discrepancy when the enzyme level was<800U/L ( P >0 05), but BGH ALT diagnosis showed discrepancy although the discrepancy was not very significant (0 01< P <0 05). When ALT>800U/L, all diagnosis kit showed significance discrepancy ( P <0 01). Measurement of ALT in the diluted sera with high concentration of enzyme showed no significance discrepancy ( P >0 05). Conclusion Suitable diagnosis kit should be chosen for the measurement of ALT level. The enzymetic activity should be dtermined within the linear range of the standard curve. Other parameters like setting the reagent blank, dynamic blank and initial absorbance should also be taken into account in order to get consistant and accurate results.
