摘要
在传统分批发酵工艺的基础上,对E.Coli表达IL-6的补料分批发酵工艺进行研究,结果表明:在发酵诱导前补料可在表达纯度相差无几的情况下提高菌体的产量,最终提高了生产效率,降低了成本。通过几次发酵比较,结果如下:分批补料工艺菌体生物量平均为10.8g/L,SDS-PAGE电泳法测IL-6表达量平均为40%,比分批发酵8.3g/L的菌体生物量及3.5%的纯度提高30.1%和3.9%。
Based on the tradional batch fermentation technology,the supplement batch fermentation technology of IL-6 expressed by E.coli is studied.The results indicates that during inducing the supplemented medium increases the bacterium number when the difference in expression purity is negligible,finally increases production efficiency and reduces production costs.By new technology,at the end of culture the average of bacterium number reached 10.8 g/L,the average of expression capacity for IL-6 was 40% by SDS-PAGE.compared with previous technology which bacterium number was 8.3 g/L and expression capacity was 38.5%,the bacterium number was increased by 30.1%,the expression purity was increased by 3.9%.
