摘要
目的 观察肝缺血再灌注后血及胃黏膜中TNF和IL 8浓度的变化及mRNA的原位表达 ,以探讨其与胃黏膜损伤的关系。方法 13 0只SD大鼠 ,制备肝脏再灌注模型 ,随机分为 3组 :( 1)假手术组 ,术中只牵拉分离肝十二指肠韧带。 ( 2 )缺血再灌注组 ,分为肝脏缺血 2 0min和 40min 2组 ,再灌注 1h ,2 4h ,72h组。 ( 3 )肝缺血再灌注加法莫替丁治疗组 ( 0 .2mg/10 0 g)。术后测定ALT ,AST各组留取胃黏膜行光镜及电镜检查 ,测定血和胃黏膜组织中肿瘤坏死因子 (TNF ) ,IL 8浓度的变化 ;利用原位分子杂交技术检测TNF ,IL 8在胃黏膜中的表达。结果 缺血再灌注后 ,胃黏膜出现炎症性损伤 ,血及胃黏膜中TNF ,IL 8浓度升高 ( P <0 .0 1) ,胃黏膜中TNF ,IL 8mRNA有不同程度的表达。结论 TNF和IL 8浓度升高及其mRNA在胃黏膜中的阳性表达 ,可能是胃黏膜的损伤因素之一。
Objective To investigate the changes of the levels of TNF and IL 8 in blood and in gastric mucosa and their mRNA expression after liver ischemia reperfusion (I/R). Methods 130 rats were randomly divided into 3 groups: (1) sham operation group(control group, n=10);(2)liver ischemia reperfusion group (n=60), including ischemia for 20min and 40min, and reperfusion for 1h, 24h and 72h respectively; (3) liver I/R rats treated with famotidine group (n=60). ALT and AST were detected after operation, pathomorphological changes of gastric mucosa were studied by light microscope and transmission electronic microscope, the concentrations of TNF and IL 8 in blood and gastric mucosa were measured by radioimmunoassay (RIA), TNF mRNA and IL 8 mRNA were detected by in situ hybridization. Results After liver I/R,inflammatory damage appeared in gastric mucosa, TNF and IL 8 concertrations increased significantly(P<0.01); TNF mRNA and IL 8 mRNA were detected in mucosa . Conclusions Increased levels of TNF and IL 8 and their mRNA expression may be the main reason in the damage of gastric mucosa in liver I/R.
