摘要
采用鲁米诺(Luminol)依赖性化学发光(CL)法,对人外周血淋巴细胞(PBL)活化早期氧自由基(OR)的生成进行了研究。结果表明,PBL 在ConA、PHA、SPA 等诱导下均可迅速产生OR,但不同刺激剂诱导OR 生成的强度及动力学不完全相同。通过进一步观察各种OR 清除剂对PBL—CL 的效应及过氧化氢酶对H_2O_2—Luminol 发光体系的影响,证明淋巴细胞在活化早期可产生各种类型的OR,包括OH、(?)OH、(?)O_2、O_2^-及H_2HO_2。
The generation of oxygen radicals(OR) by human peripheral bloodIymphocytes(PBL)had been studied using a luminol-dependent chemiluminescence(CL).The results showed that PBL might generate OR immediately after stimu-lated with various mitogens such as ConA,PHA and SPA.However,LPS(6.25—400ug/ml)was unable to mediate such an effect.In addition,specific scavengersof OR were used to investigate their effects on hTman PBL—CL and determinedthe kinds of OR released by PBL stimulated with ConA,PHA and SPA.CL res-ponse of PBL may be significantly suppressed by sodium azide('O_2 scavenger),sodivm benzoate('OH scavenger)and SOD (O_2 seavenger),whereas eatalase(H_20_2scauenger)showed an enhancinng effect on the CL.The data suggested that thestimulation of human PBL with mitogens results in an oxidative burst withthe liberation of oxygen radicals,including 'O_2,~.OH,O_2^-. and H_2O_2
